摘要
目的建立高效液相色谱法(HPLC法)测定氧氟沙星滴眼液中抑菌剂羟苯乙酯的含量。方法选择Agilent ZORBAX SB-Aq (C_(18),4.6mm×250mm,5μm),以醋酸铵高氯酸钠溶液(取醋酸铵4.0g和高氯酸钠7.0g,加水1300mL使溶解,用磷酸调节pH值至2.2)-乙腈(85∶15)为流动相A,乙腈为流动相B,按线性梯度洗脱,流速为1.0mL·min^(-1),柱温为40℃,检测波长为256nm。结果氧氟沙星滴眼液中羟苯乙酯回归方程为y=44902090x-10297.8(R=0.9999),在0.0245~0.1837mg·mL^(-1)范围内线性关系良好;结果平均回收率为100.28%,RSD为0.92%(n=9)。结论该方法简便、准确、专属性强、重现性良好,适用于氧氟沙星滴眼液中抑菌剂羟苯乙酯的含量测定。
OBJECTIVE To establish a HPLC method for the determination of the content of ethylparaben in Ofloxacin Eye Drops.METHODS The mobile phase consisted of ammonium acetate and sodium perchlorate solution(dissolve 4.0 g ammonium acetate and 7.0 g sodium perchlorate to 1300 mL with water,adjusted pH to 2.2 with phosphoric acid)-acetonitrile(85∶15)(A) and acetonitrile(B) with gradient elution.The flow rate was 1.0 mL·min^-1,and the temperature of column was 40℃.The detection wavelength was 256 nm,.RESULTS The regress equation for ethylparaben was Y=44902090 x^-10297.8(R=0.9999),and the linear range was 0.0245-0.1837 mg·mL^-1.The average recovery of ethylparaben was 100.28%,RSD=0.92%(n=9).CONCLUSION The method is simple,fast,and accurate,with high sensitivity and good repeatability,and can be used for content determination of ethylparaben.
