CK18在乳腺癌血清中的表达与临床意义

Expression of serum cytokeratin-18 in breast cancer and its clinical significance

目的:根据CK18在乳腺癌患者血清中的表达情况,分析其表达的临床意义,探讨CK18作为辅助诊断乳腺癌分子生物标志物的可行性。方法:随机选取2016年9～12月病理诊断为乳腺癌的患者血清和健康人的血清各26例,采用蛋白质免疫印迹方法检测CK18的表达情况,并分析其临床意义,同时比较血清中CK18与各临床因素之间的关系。结果:在26例健康血清中,CK18阳性表达25例,阳性率96%;在26例乳腺癌患者血清中,CK18阳性率表达19例,阳性率73%。比较两组血清中CK18和GAPDH相对灰度比值,乳腺癌患者血清中CK18表达高于健康人,差异有统计学意义(P <0.05),比较其CK18表达和各临床因素的关系,显示差异无统计学意义(P> 0.05)。结论:和正常人血清相比,CK18在乳腺癌血清中高表达,这可能对乳腺癌的早期诊断、疾病进展、评价生存和预后具有重要的意义。另外在乳腺癌患者血清中CK18的表达与年龄、肿瘤大小、WHO分级高低、病理类型、ER、PR、HER-2表达与否均无明显的相关性。

Objective: To determine the expression of serum cytokeratin-18 (CK18) in patients with breast cancer and analyze its clinical significance, and to investigate the feasibility of using CK18 as a molecular biomarker for breast cancer diagnosis. Methods: Serum samples were randomly collected from 26 patients pathologically diagnosed with breast cancer from September to November, 2017, as well as 26 healthy people. Western blot was used to determine the expression of CK18, and its clinical significance was analyzed. Meanwhile, the relationship between CK18 and various clinical factors was evaluated. Results:It was found that CK18 was positively expressed in 25 cases in healthy serum, and the positive rate was 96%. In 26 cases of breast cancer patients, 19 cases was positive, which rate was 73%.The CK18/GAPDH grayscale ratio in serum was compared between the two groups. The expression of serum CK18 in breast cancer patients was significantly higher than that in healthy people (P < 0.05). There was no significant relationship between the expression of CK18 and clinical factors (P > 0.05). Conclusion: Compared with healthy serum, CK18 was highly expressed in breast cancer serum. which may be important for the early diagnosis, disease progression, and survival and prognosis evaluation of breast cancer. In addition, the expression of serum CK18 is not associated with age, tumor size, WHO grade, pathological type, and ER, PR, or HER-2 expression in patients with breast cancer.
