摘要
为研究姜黄素对单侧输尿管阻塞大鼠肾间质纤维化可能的干预机制。将SD大鼠随机分为假手术组(Sham)、对照组(UUO)、UUO+50 mg·kg^(-1)姜黄素组,UUO+75 mg·kg^(-1)姜黄素组,每组6只,均于手术当天晚上灌胃给药,每24 h给药1次。第14天处死取肾组织,Masson染色光学显微镜下肾脏组织病理变化、试剂盒检测肾组织匀浆液丙二醛(MDA)含量,谷胱甘肽S转移酶(GSH)、超氧化物歧化酶(SOD)和过氧化氢酶(CAT)活力;RT-PCR法检测核转录相关因子2(Nrf2)、血红素加氧酶(HO-1)mRNA表达。结果表明:与Sham组比较,UUO组和姜黄素干预组MDA含量均升高,GSH、SOD、CAT活力均显著降低,Nrf2和HO-1 mRNA表达显著升高;UUO组与姜黄素干预组比较,姜黄素干预组MDA含量显著降低,GSH、SOD、CAT显著升高,Nrf2和HO-1 mRNA表达显著升高;UUO组肾小球球囊间隙增大,排列紊乱,胶原纤维增多,明显纤维化变性,姜黄素干预组病理表现较UUO组明显减轻。姜黄素能减轻单侧输尿管梗阻大鼠肾脏间质纤维化,其机制可能是通过诱导肾组织Nrf2表达,上调HO-1、SOD等抗氧化酶水平,提高肾组织抗氧化应激水平。
To investigate the intervention mechanism of curcumin on renal interstitial fibrosis in rats with unilateral ureteral obstruction,SD rats were randomly divided into Sham group,UUO group,UUO+50 mg ·kg^-1 curcumin group,UUO+75 mg ·kg^-1 curcumin group,6 rats in each group,they were administered by intragastric administration on the evening of the operation,once every 24 h.Kidney tissue was sacrificed on the 14th day,pathological changes of kidney tissue under Masson staining light microscope,kit for detection of malondialdehyde (MDA) content in kidney homogenate,glutathione S transferase (GSH),superoxide dismutase(SOD) and catalase (CAT) activity;RT-PCR was used to detect nuclear transcription-related factor 2 (Nrf2) and heme oxygenase (HO-1) mRNA expression.The results showed that compared with Sham group,MDA content in UUO group and curcumin group increased significantly,GSH,SOD,CAT activity decreased significantly,Nrf2 and HO-1 mRNA expression increased significantly;UUO group and curcumin intervention group,turmeric the MDA content in the intervention group was significantly decreased,GSH,SOD and CAT were significantly increased,and the expression of Nrf2 and HO-1 mRNA was significantly increased.In the UUO group,the glomerular balloon space was enlarged,the arrangement was disordered,collagen fibers were increased,and fibrosis was obvious.The pathological manifestations of the curcumin intervention group were significantly less than those of the UUO group.Curcumin can alleviate renal interstitial fibrosis in rats with unilateral ureteral obstruction.The mechanism may be through inducing Nrf2 expression in kidney tissue,up-regulating the levels of antioxidant enzymes such as HO-1 and SOD,and increasing the level of anti-oxidative stress in renal tissues.
