川射干的超声-酶法辅助半仿生提取工艺优选 被引量:3

Optimization of ultrasonic wave-enzymatic assisted semi-bionic extraction technology for Iris tectorum Maxim

摘要 应用超声-酶法辅助半仿生,以总异黄酮得率和干膏收率为评价指标,通过正交实验考察超声功率、超声时间及溶剂用量3个因素对川射干半仿生超声提取工艺的影响。采用紫外分光光度法测定总异黄酮含量,检测波长266 nm。最佳提取工艺为复合酶预处理后于200 W超声提取,依次加入液料比[V(缓冲液)∶m(川射干药粉)]为15∶1 mL/g的pH=2.2、7.4、8.0磷酸氢二钠-柠檬酸缓冲液提取3次,每次15 min;总异黄酮得率为6.92%(RSD为1.45%),干膏收率为42.18%(RSD为1.21%)。该法总异黄酮得率和干膏收率高于乙醇回流法,其工艺参数可为药材工业生产提供参考。 Ultrasonic wave-enzymatic assisted semi-biotic extraction was used to evaluate the yield of total isoflavones and dry extract.Orthogonal test was adopted to optimize extraction technology by taking ultrasonic power,extraction time and solvent amount as factors.UV was adopted to determine the content of total isoflavones at 266 nm.Optimal extraction process of Iris tectorum Maxim.was as follows:pretreatment by cellulase-pectinase,ultrasonic power of 200 W,extracted thrice by sodium hydrogen phosphate-citric acid buffer with pH 2.2,7.4 and 8.0 successively, and liquid-solid ratio was 15∶1 mL/g every time,extraction time of 15 min,respectively.Yields of total isoflavones and dry extract were 6.92% and 42.18% with RSD of 1.45 % and 1.21%,respectively.Yields of total isoflavones and dry extract of this optimized process were higher than ethanol reflux process.This study could provide a reference for industrial production of Iris tectorum Maxim.
