Expression of Twist and matrix metalloproteinase-2 in pancreatic cancer tissues and its clinical significance

目的研究Twist和基质金属蛋白酶(MMP)-2在胰腺癌组织和正常胰腺组织中的表达情况,研究二种因子与胰腺癌患者的年龄、性别、肿瘤部位、病理分级、临床分期和淋巴结转移的关系及临床意义。方法采用Elivision Plus法对48例手术切除的胰腺癌组织和20例癌旁正常胰腺组织中Twist和MMP-2的表达进行检测,结合临床病理特点进行分析。结果 Twist、MMP-2蛋白在胰腺癌中阳性表达率为67%、75%,明显高于其在正常组织中的阳性表达率20%、35%,差异有统计学意义(χ2值分别为12.341和9.716,P<0.01)。胰腺癌组织中Twist与MMP-2的阳性表达呈正相关(r=0.272,P<0.05)。结论胰腺癌癌组织中Twist和MMP-2呈高表达,且两者呈正相关,联合检测二者在胰腺癌中的阳性表达,有助于研究胰腺癌的发生、发展。二者在癌组织中的表达明显高于正常胰腺组织,且与癌组织的分化程度、临床分期及淋巴结转移有关,随着胰腺癌临床分期和组织分化程度进展及淋巴结转移而增高。表明二者可能与胰腺癌浸润、转移及生长有关。

Objective To investigate the expression of Twist and matrix metalloproteinase-2(MMP-2) in pancreatic cancer tissues and normal pancreatic tissues, and to determine relationship between the two factors and age,gender, tumor site, pathological grade, clinical stage and lymph node metastasis and its clinical significance. Methods Elivision Plus method was used to determine the expression levels of Twist and MMP-2 in 48 surgically resected pancreatic cancer tissues and 20 normal adjacent pancreatic tissues. The clinical and pathological features were analyzed. Results The positive expression rates of Twist and MMP-2 in pancreatic cancer tissues were significantly higher than those in normal adjacent tissues(67% vs 20%;75% vs 35%), with statistically significant differences(both P<0.01). There was a positive correlation between Twist and MMP-2 expression in pancreatic cancer tissues(r=0.272, P<0.05). Conclusion Twist and MMP-2 are highly expressed in pancreatic cancer tissues, and they are positively correlated. The combined examination of their positive expression in pancreatic cancer favors the study on occurrence and development of pancreatic cancer. The expression levels of Twist and MMP-2 in pancreatic cancer tissues are significantly higher than those in normal pancreatic tissues, and are related to the degree of differentiation,clinical stage and lymph node metastasis of pancreatic cancer tissues. The levels increase along with the clinical progression, tissue differentiation and lymph node metastasis in pancreatic cancer, which may be related to the invasion,metastasis and growth of pancreatic cancer.
