红景天苷对人肾癌细胞生长、侵袭和迁移的调节作用

Effects of salidroside on cell growth, invasion and migration in human renal cell carcinoma cells

目的探究红景天苷(SD)对人肾癌细胞生长、侵袭和迁移的作用。方法以0、2、5、10μmol/L SD作用于体外培养的人肾癌细胞Ketr-3,利用CCK-8法检测培养不同时间(0、1、2、3、4 d)细胞的增殖情况,用Transwell实验检测细胞侵袭能力,用划痕实验检测细胞迁移能力,用Western blot检测Ki67、血管内皮生长因子(VEGF)、基质金属酶蛋白-9 (MMP-9)蛋白表达水平。结果 SD浓度达到20μmol/L及以上时,Ketr-3细胞活性降低;SD (2、5、10μmol/L)作用4 d后,与Control组比较,SD (2、5、10μmol/L)组Ketr-3和Caki-1细胞增殖倍数显著降低(P<0.01),Ki67的表达水平显著降低(P<0.01);SD(2、5、10μmol/L)作用24 h后,Ketr-3和Caki-1细胞侵袭能力明显减弱(P<0.01),与Control组相比,SD(5、10μmol/L)组迁移能力均显著减弱(P<0.01),VEGF和MMP-9的表达水平显著降低(P<0.01)。结论 SD能够抑制人肾癌细胞Ketr-3和Caki-1的生长、侵袭和迁移。

Objective To investigate the effects of salidroside(SD)on cell growth,invasion and migration in human renal cell carcinoma cells.Methods After treated with 0,2,5,10μmol/L SD for 0,1,2,3,4 days,the rate of cell growth was detected by CCK-8 assay.Transwell assay was used to investigate the effects of SD on cell invasion.Wound healing assay was used to investigate the effects of SD on cell migration.The effects of SD on protein expressions of Ki67,VEGF,MMP-9 were determined by Western blot assay.Results After treated with SD(>20μmol/L),cell viability was declined compared with Control group.After treated with SD(2,5,10μmol/L)for 4 days,cell growth(folds)of Ketr-3 and Caki-1 cells was decreased significantly compared with group Control(P<0.01).Protein expression of Ki67 was decreased significantly compared with Control group(P<0.01).After treated with SD(2,5,10μmol/L)for 24 h,the ability of cell invasion of Ketr-3 and Caki-1 cells was weakened(P<0.01).Compared with Control group,the ability of migration in SD(5,10μmol/L)group were decreased significantly(P<0.01).Protein expressions of VEGF and MMP-9 were decreased significantly compared with Control group(P<0.01).Conclusion Salidroside can inhibit cell growth,invasion and migration in human renal cell carcinoma Ketr-3 and Caki-1 cells.
