摘要
目的探讨侧脑室注射霍乱毒素B亚单位(CTB)对小鼠免疫细胞及细胞因子的影响。方法 C57雌性小鼠侧脑室注射CTB,运用流式细胞术检测外周血免疫细胞亚群变化;流式微球捕获芯片技术(CBA)检测血清中细胞因子及趋化因子的变化。结果 CTB能够明显升高单核巨噬细胞、CD4^+T细胞及B细胞的比例;炎症因子γ干扰素(IFN-γ)、肿瘤坏死因子α(TNF-α)、白细胞介素-6(IL-6)、白细胞介素-9(IL-9)及趋化因子巨噬细胞炎性蛋白-1α(MIP-1α)、巨噬细胞炎性蛋白-1β(MIP-1β)、B淋巴细胞趋化因子(BLC)、γ干扰素诱导的单核因子(Mig)、调节活化正常T细胞表达与分泌的趋化因子(RANTES)、单核细胞趋化蛋白-1(MCP-1)也相应升高。结论侧脑室注射CTB能够明显改变小鼠免疫应答状态。
Objective To explore the effect of cholera toxin subunit B on the immune system of mice.Methods Intra-lateral ventricle(LV)of C57 female mice were injected with toxin subunit B.Immunocyte in peripheral blood were detected by flow cytometry;Cytokine and chemokines in serum were detected by flow cytometry with Cytometrin Bead Array(CBA).Results Data showed that CD3^+CD4^+T cells,B cells and mononuclear macrophage were higher than PBS control group.IFN-γ,TNF-α,IL-6,IL-9 inflammation cytokines and MIP-1α,MIP-1β,BLC,MIG,RANTES,MCP1 chemokine were higher than PBS control group.Conclusion Cholera toxin subunit B can obviously change the state of immune response in mice.
