艾附暖宫软胶囊镇痛抗炎作用研究及对雌性幼小鼠和催产素所致小鼠痛经模型的影响

Anti-Inflammatory and Analgesic Effects of Aifu Nuangong Soft Capsule and Its Influence to Dysmenorrhea Models Induced by Oxytocin

目的:研究艾附暖宫软胶囊的镇痛抗炎作用,对雌性幼小鼠生殖器官发育的影响以及对雌二醇含量的影响,观察对痛经模型小鼠的作用。方法:采用小鼠醋酸扭体法及热板法疼痛模型,观察艾附暖宫软胶囊的镇痛作用,二甲苯致小鼠耳肿胀法、腹腔染料渗出法等炎症模型观察艾附暖宫软胶囊的抗炎作用;采用未成年小鼠,观察药物对小鼠子宫、卵巢的影响,并采用酶联免疫法(ELISA法)检测小鼠血清中雌二醇(E2)的含量;采用催产素致小鼠痛经模型,观察小鼠疼痛扭体反应的次数。结果:艾附暖宫软胶囊能减少高剂量组醋酸所致小鼠扭体次数(P<0.05),延长小鼠热刺激痛阈时间(P<0.01);显著抑制二甲苯所致小鼠耳肿胀度,降低肿胀率,高剂量组显著降低腹腔染料渗出量(P<0.01),有明显抑制炎性渗出的作用;与阴性对照组相比,艾附暖宫软胶囊增加卵巢系数和子宫系数,各剂量组血清雌二醇含量显著增加。结论:艾附暖宫软胶囊具有良好的镇痛抗炎作用,能促进未成年小鼠生殖器官的发育,增加血清雌二醇含量,表明该药具有类似雌激素样作用。

Objective: To investigate the anti-inflammatory and analgesic effects of Aifu Nuangong Soft Capsule, to explore its influences to the reproductive organs of female immature mice and its effect on the serum content of E2, and to observe its effect on dysmenorrhea models. Methods: The analgesic effect of Aifu Nuangong Soft Capsule was observed by acetic acid writhing test and hot plate test;its anti-inflammatory effect was observed by ear swelling model induced by xylene and the coeliac dye exudation model. For immature mice, uterus and ovaries were weighted and calculated, the content of E2 was detected by ELISA method. The model of dysmenorrhea was duplicated with oxytocin in mice, and the effects of different doses of Aifu Nuangong Soft Capsule on the analgesic effect in mice were observed through writhing test. Results: After the intervention of Aifu Nuangong Soft Capsule of high-dose, the times of writhing induced by acetic acid was significantly reduced( P <0.05);pain thresholds was significantly increased in the hot plate test( P <0.01);ear swelling was inhibited, the swelling rate was lowered, and dye exudation was significantly decreased( P <0.01). Coefficients of uterus and ovary, as well as the serum level of E2 were increased in all medication groups( P <0.01). Conclusion: Aifu Nuangong Soft Capsule has better analgesic and anti-inflammatory effects, it can promote the development of reproductive organs in immature mice, it also can increase the serum level of E2, which shows that it has estrogen-like effect.
