摘要
目的:探讨苦参素(OMT)预处理对肾缺血再灌注(I/R)损伤的保护作用及核转录因子-κB表达的影响。方法:将SD大鼠随机分为假手术组(S组)、I/R组、OMT高、低剂量组(H、L组),每组10只。H、L组在I/R造模前进行OMT给药干预,S、I/R组给予等体积0.9%氯化钠注射液处理。检测并比较各组动物肾损伤指标、炎性因子水平、氧化应激指标水平差异以及肾脏组织中核转录因子-κB(NF-κB p65)蛋白表达水平变化。结果:与S组比较,I/R组肾组织损伤明显,肌酐(Cr)、血尿素氮(BUN)、白细胞介素-1β(IL-1β)、白细胞介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)及丙二醛(MDA)水平均明显升高,超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)活性明显降低,NF-κB p65蛋白表达水平明显升高(P<0.05)。与I/R组比较,OMT预处理组肾小管损伤明显减轻,Cr、BUN、IL-1β、IL-6、TNF-α及MDA水平均明显降低,SOD和GSH-Px活性明显升高,NF-κB p65蛋白表达水平明显降低,且H组效果更明显(P<0.05)。结论:苦参素预处理对肾脏有保护作用,其机制可能与增强肾脏抗氧化能力,减轻炎症反应,抑制NF-κB p65表达有关。
Objective:To investigate the protective effect of oxymatrine preconditioning on renal ischemia/reperfusion injury and expression of nuclear factor-κB.Methods:SD rats were randomly divided into sham operation group(S group),I/R group,OMT high and low dose group(H,L group),with 10 rats in each group.H and L group were treated with oxymatrine before modeling,S and I/R group were given 0.9%sodium chloride injection of the same volume.The levels of renal injury indexes,inflammatory factors,oxidative stress indexes and nuclear factor-κB(NF-κB p65)protein expression in each group were detected and compared.Results:Compared with S group,renal tissue injury was obvious in I/R group.Meanwhile,the levels of creatinine(Cr),blood urea nitrogen(BUN),interleukin-1β(IL-1β),interleukin-6(IL-6),tumor necrosis factor-α(TNF-α)and malondialdehyde(MDA)were significantly increased,superoxide dismutase(SOD)and glutathione peroxidase(GSH-Px)activity was significantly decreased,and the expression of NF-κB p65 protein was significantly increased in I/R group(P<0.05).Compared with I/R group,the renal tubular injury was significantly alleviated in the oxymatrine preconditioning group.And the levels of Cr,BUN,IL-1β,IL-6,TNF-αand MDA were significantly decreased,the activities of SOD and GSH-Px were significantly increased,the expression of NF-κB p65 protein was significantly decreased in the Oxymatrine preconditioning group,and the effect of H group was more obvious(P<0.05).Conclusion:Oxymatrine preconditioning has a protective effect on kidney,and its mechanism may be related to enhancing renal antioxidant capacity,alleviating inflammation and inhibiting the expression of NF-κB p65 protein.
