摘要
目的:探讨电针介导下MAPK/ERK信号通路参与分娩镇痛的作用机制。方法:将120例分娩模型大鼠按照随机数字表法分为瑞芬太尼组、电针三阴交加合谷组、电针夹脊穴组及空白对照组各30例。应用热水甩尾检测痛阈,Western blot及Realtime PCR检测大鼠中枢组织Raf、ERK、CREB及子宫组织DYN蛋白与基因表达。结果:治疗前4组大鼠痛阈值组间比较差异无统计学意义,治疗后组间比较差异有统计学意义; Raf、CREB、ERK蛋白与基因在大脑灰质表达组间比较差异无统计学意义,与脊髓膨大段组间比较差异有统计学意义,DYN基因与蛋白在子宫表达组间比较差异有统计学意义。结论:电针夹脊穴组、电针三阴交加合谷组能提高分娩大鼠痛阈及降低其脊髓Raf/ERK/CREB与提高其子宫DYN基因与蛋白表达水平。
Purpose:To investigate the mechanism of electroacupuncture-mediated MAPK/ERK signaling involved in labor analgesia.Methods:For 120 cases of childbirth model rats,30 cases in each group were randomly divided into remifentanil group,electroacupuncture Sanyinjiao plus Hegu group,electro-acupuncture Jiaji acupoint group and blank control group.Using hot water tail flick to detect pain threshold,Western blot and Real-time PCR detection of Raf,ERK,CREB and DYN protein and gene expression in rat central tissue Results:Before treatment,there was no significant difference in pain threshold between the four groups.After treatment,the difference between the groups was statistically significant;There was no significant difference in protein expression between gray matter expression group and cerebral gray matter.DYN gene and protein were expressed in sub-government groups.There was a significant difference between groups.Conclusion:Electroacupuncture at Jiaji acupoint group and electroacupuncture at Sanyinjiao plus Hegu group can improve the pain threshold of rats at delivery,reduce their spinal cord Raf/ERK/CREB,and increase their daughter DYN gene.
