麝香通心滴丸对动脉粥样硬化模型大鼠颈总动脉形态及hs-CRP的影响 被引量:2

Effects of Shexiang Tongxin Dropping Pills on the Morphology of Common Carotid Artery and hs-CRP in AS Model Rats

摘要 目的观察麝香通心滴丸对动脉粥样硬化大鼠模型超敏C反应蛋白(hs-CRP)、血清脂蛋白相关磷脂酶A2(Lp-PL-A2)的影响,以及动脉粥样硬化大鼠模型颈总动脉的形态学变化。探讨麝香通心滴丸可能的作用机制,为临床治疗稳定型心绞痛提供依据。方法选择健康雄性SD大鼠60只,随机取15只作为空白对照组,剩下的45只采取高脂饮食与注射垂体后叶素方法诱导大鼠发生动脉粥样硬化,建立动脉粥样硬化动物模型。将造模成功的30只大鼠随机分为3组,模型对照组(同体积蒸馏水每日1次)、中药治疗组[麝香通心滴丸溶液10.94 mg/(kg·d)]、西药对照组[瑞舒伐他汀溶液0.52 mg/(kg·d)],每组各10只,空白对照组也予以同体积蒸馏水,每日1次,分别灌胃8周后,乳胶凝集比浊法测定各组大鼠hs-CRP含量,试剂盒测定各组Lp-PL-A2的含量,观察各组大鼠血清中hs-CRP、Lp-PL-A2变化;取大鼠颈总动脉,光镜下观察形态学变化。结果 1)与模型对照组相比,空白对照组、中药治疗组、西药对照组的hs-CRP水平明显降低,差异有统计学意义(P <0.05);与西药对照组相比,中药治疗组大鼠hs-CRP水平差异无统计学意义(P> 0.05)。2)与模型对照组相比,空白对照组、中药治疗组、西药对照组的Lp-PL-A2水平明显降低,差异有统计学意义(P <0.05);与西药对照组相比,中药治疗组大鼠Lp-PL-A2水平差异无统计学意义(P> 0.05)。3)与空白对照组相比,模型对照组、中药治疗组、西药对照组大鼠颈总动脉病理改变明显;与模型对照组相比,中药治疗组大鼠颈总动脉组织病理改变减轻。结论 1)麝香通心滴丸可降低动脉粥样硬化大鼠hs-CRP、Lp-PL-A2含量,这可能是其抗动脉粥样硬化的机制之一。2)麝香通心滴丸可明显改善动脉粥样硬化大鼠颈总动脉病理形态学变化。 Objective: To observe the effects of Shexiang Tongxin Dropping Pills on hypersensitive C-reactive protein,serum lipoprotein-related phospholipase A2 in atherosclerotic rat model and the morphological changes of common carotid artery in atherosclerotic rat model,and to explore its possible mechanism and provide basis for clinical treatment of stable angina pectoris. Methods: 60 healthy male SD rats were randomly selected as the blank group. The remaining 45 rats were induced to develop atherosclerosis by high-fat diet and injection of pituitrin. The Atherosclerosis Animal model was established. 30 rats were randomly divided into three groups: the model group(once a day with distilled water of the same volume),TCM group [10.94 mg/(kg·d) and WM group(0.52 mg/(kg·d)],10 rats in each. The blank group were given distilled water of the same volume once a day. After 8 weeks of intragastric administration,the content of hs-CRP was determined by latex agglutination turbidimetry,and the content of Lp-PL-A2 was determined by kit. The changes of hs-CRP and Lp-PL-A2 in serum were observed. The common carotid artery was taken and the morphological changes were observed under light microscope. Results: Compared with the model group,the hs-CRP and Lp-PL-A2 levels in the blank group, TCM group and WM group were significantly lower(P < 0.05);compared with WM group,those in TCM group were not significantly different(P > 0.05). Compared with the blank group,the pathological changes of the common carotid artery in the model group,TCM group and WM group were obvious;compared with the model group,those in TCM group were alleviated. Conclusion: Shexiang Tongxin Dropping Pills can reduce the contents of hs-CRP and Lp-PL-A2 in atherosclerotic rats,which may be one of its anti-AS mechanisms. Shexiang Tongxin Dropping Pills can significantly improve the pathomorphological changes of common carotid artery in atherosclerotic rats.
