摘要
以新鲜的百香果果肉为原料,用PCA培养基对百香果果肉中的内生细菌进行分离纯化,然后用16S rDNA测序法鉴定菌种,最后测定菌株的生长条件。菌种分离结果显示,从百香果果肉分离出了3类菌;菌种鉴定结果显示,白色菌落为Enterobactersp.,黄色菌落为Pseudomonas sp.;菌种生长条件结果显示,3株受试菌均在LB培养基中生长最好,在pH 5.0~8.0具有良好的生长状况,最适温度约为35℃。
Using fresh passion sarcocarp as raw material, the endophytic bacteria in Passion sarcocarp were isolated and purified by PCA medium. Three strains of characteristic bacteria were identified by 16 S rDNA sequence analysis method, and the optimum growth conditions were determined. The results showed that the single colony morphology of endophytic bacteria isolated from passion sarcocarp was white or yellow, its surface was raised and smooth, opaque and mucous. The 16 S rDNA analysis of three strains of characteristic bacteria showed that they belong to Enterobacter sp. or Pseudomonas sp. Three strains grew well in LB medium;the optimum growth temperature was 35 ℃;the range of favourable pH value is 5.0~8.0.
