摘要
目的:探讨使用不同试剂对病理组织进行苏木精-伊红染色(HE)染色处理的效果。方法:选取使用传统试剂已经进行过HE染色处理的100例片子纳入对照组,选取使用环保试剂进行HE染色处理的100例片子纳入观察组。分析对比两组病理组织的染色色彩、对比度、色彩渗透力、呈现图像的速度及图像结构清晰程度;比较相同病理组织在不同染色试剂下的HE染色效果。结果:观察组各种病理组织的HE染色效果色彩较为鲜艳,但两组在Ⅰ级、Ⅱ级、Ⅲ级切片中的色彩比较,差异均无统计学意义(x2=0.604,x2=0.533,x2=0.785;P>0.05);两组HE染色的对比度、色彩渗透力、图像呈现速度和图像结构清晰程度比较,差异均无统计学意义[对比度(x2=0.324,x2=0.001,x2=0.546;P>0.05);色彩渗透力(x2=0.529,x2=0.001,x2=0.631;P>0.05);图像呈现速度(x2=0.312,x2=0.268,x2=0.721;P>0.05);图像结构清晰程度(x2=0.692,x2=279,x2=0.301;P>0.05)]。结论:新型环保试剂应用于病理组织的HE染色,其效果与传统试剂一致,同时具有环保、无毒、无害等特点,值得推广使用。
Objective: To investigate the effect of hematoxylin-eosin staining (HE) with different reagents on pathological tissues. Methods: 100 sections which had been dyed by HE with traditional reagents were selected as control group, and 100 sections which had been dyed by HE with environmental protection reagents were selected as observation group. The dyeing color, contrast, color penetration, speed of presenting image and clarity of image structure between two groups were analyzed and compared. And HE staining effects of the same pathological tissue under different dyeing reagents were compared. Results: The HE staining effects of various pathological tissues of the observation group was brighter, but there were no significant differences between the two groups in grade I, II and III sections (x^2=0.604, x^2=0.533, x^2=0.785, P>0.05). There were no significant differences in contrast, color penetration, speed of image presentation and clarity of image structure between the two groups [contrast (x^2=0.324, x^2=0.001, x^2=0.546, P>0.05), color penetration (x^2=0.529, x^2=0.001, x^2=0.631, P>0.05), speed of image presentation (x^2=0.312, x^2=0.268, x^2=0.721, P>0.05), clarity of image structure (x^2=0.692, x^2=279, x^2=0.301, P>0.05)]. Conclusion: The new type of environment protection reagent that is used in HE staining of pathological tissues has the same effect with the traditional reagent, meanwhile it has the characteristics include environmental protection, non-toxicity and harmlessness. It is worth to be popularized and used.
