摘要
目的:观察欣力康胶囊对尾静脉注射肺癌细胞建立的肺转移模型小鼠生存期和生存状态的影响,并分析其潜在的作用机制。方法:尾静脉注射造模,建立Lewis 肺癌(lewis lung carcinoma, LLC)转移模型小鼠,共分3 组,每组各10 只,分别为生理盐水组、低浓度欣力康组(0.08 g/mL)、高浓度欣力康组(0.40 g/mL)。分别观察3 组小鼠精神状态、摄食量、毛色光泽度以及生存期,同时荧光素酶报告基因法(Luciferase assay)分析欣力康胶囊对肿瘤相关信号转导与激活因子3(signal transducer and activator of transcription 3, STAT3)和核转录因子-κB(nuclear factor-κB, NFκB)信号通路的影响。结果:生理盐水组小鼠普遍表现为精神萎靡、嗜睡,毛色无光泽,基本无活动,且极少进食与饮水;低浓度欣力康组小鼠表现与生理盐水组相似;高浓度欣力康组小鼠精神状态明显改善,毛色相对光泽,活动性增强,且进食与饮水量明显增加。生理盐水组小鼠中位生存时间为17 d,低浓度欣力康组为17.5 d,高浓度欣力康组为21 d,高浓度欣力康组小鼠生存时间明显长于生理盐水组(P <0.05)。欣力康胶囊能够显著抑制肿瘤相关的STAT3 和NFκB 信号通路。结论:欣力康胶囊能够延长肺癌转移模型小鼠的生存时间,改善模型小鼠的生存状态,这可能和欣力康抑制STAT3 和NFκB 信号通路有关。
Objective To study the effects of Xinlikang Capsule on survival time and life quality of the model mice of pulmonary cancer metastasis of lung cancer, and explore the mechanism. Methods Mouse model of pulmonary cancer metastasis of Lewis lung carcinoma (LLC) was established through tail intravenous injection, and were divided into saline group, low-dose Xinlikang group (0.08 g/mL) and high-dose Xinlikang group (0.40 g/mL), 10 mice in each group. The activity, food intake, hair glossiness and survival time were observed. Besides, the effects of Xinlikang on tumor-related signal transducer and activator of transcription 3 (STAT3) and nuclear factor-κB (NFκB) signal pathways were analyzed with luciferase assay. Results The mice of saline group showed as depression, drowsiness, low glossy hair, low activity, little food intake and drinking, which was similar to mice in the low-dose Xinlikang group, but the mice in high–dose Xinlikang group showed as good spirit, glossy hair, more activity and more food intake and drinking. The median survival time was 17 d in saline group, 17.5 d in low concentration group, and 21 d in high-dose Xinlikang group which was significantly more than that in saline group (P <0.05). Xinlikang Capsule significantly inhibited the STAT3 and NFκB signal pathways. Conclusion Xinlikang Capsule can prolong the survival time and improve the quality of life of mouse model with pulmonary cancer metastasis of lung cancer, maybe partly through inhibiting STAT3 and NFκB signal pathways.
