摘要
目的比较SCC9和CAL27细胞构建口腔癌原位异种移植瘤模型所需细胞浓度的差异。方法将30只裸鼠随机分为10组,每组3只,调节SCC9和CAL27细胞至1×106个/mL、2×106个/mL、5×106个/mL、1×107个/mL和2×107个/mL 5个不同浓度,分别于每组裸鼠舌部注入0.05 mL细胞悬液,观测裸鼠舌部成瘤情况。结果 SCC9细胞诱导裸鼠舌部成瘤最低细胞浓度为2×107个/mL,CAL27细胞诱导裸鼠舌部成瘤最低细胞浓度为1×106个/mL。结论与SCC9细胞相比,CAL27细胞诱导裸鼠舌部成瘤所需细胞浓度低,更适用于构建舌肿瘤模型。
Objective To compare the differences in cell concentration required for the establishment of orthotopic-xenograft nude-mouse models of oral cancer between SCC9 and CAL27 cells.Methods Thirty nude mice were randomly divided into 10 groups, 3 mice in each group. SCC9 and CAL27 cells were adjusted to 5 different concentrations of 1×10^7/mL、2×10^7/mL、5×10^7/mL、1×10^7/mL and 2×10^7/mL.0.05 mL of cell suspension was injected into the tongue of nude mice,and then the tumor formation of the tongue in nude mice was observed. Results SCC9 cells induced tumor formation in the tongue of nude mice at a minimum concentration of 2×10^7/ mL,and CAL27 cells induced tumor formation at a minimum concentration of 1×10^7/mL.Conclusions Compared with SCC9 cells, CAL27 cells required lower cell concentration to induce tongue tumor formation,which was more suitable for the establishment of tongue tumor model.
