菟丝子黄酮对幼年大鼠睾丸支持细胞增殖和分化的影响及机制 被引量:16

Effects and mechanism of flavonoid of semen cuscutae on proliferation and differentiation of sertoli cell in young rats

摘要 目的观察不同浓度菟丝子黄酮对幼年大鼠睾丸支持细胞增殖与分化的作用并探讨其机制。方法分离20 d龄幼年大鼠睾丸支持细胞,并体外培养。随机分为对照组和菟丝子黄酮低、中、高剂量(200、400、800 mg/L)组。MTT法检测各组支持细胞的增殖率;免疫组化法检测各组支持细胞中Ki67的表达;Western blot法检测各组支持细胞中蛋白CK-18、AR、p-Akt和Akt的表达水平。结果与对照组相比,菟丝子黄酮低、中、高剂量组支持细胞增殖率显著增加(P<0.01);Ki67阳性细胞数量显著增多(P<0.01);AR和p-Akt的相对表达量也显著增加(P<0.01),而CK-18的表达量显著降低(P<0.01),且有剂量依赖性(P<0.01或P<0.05)。结论菟丝子黄酮可通过上调AR的表达,激活PI3K/Akt通路,增加Akt的磷酸化,促进幼年支持细胞的增殖和分化。 Objective To investigate the effects and mechanism of flavonoid of semen cuscutae(FSC) on proliferation and differentiation of Sertoli cells in young rats.Methods Sertoli cell were cultured in vitro after separated from testes of 20 days SD rats.And they were randomly divided into four groups:control group,low-dose(200 mg/L) FSC group,mid-dose(400 mg/L) FSC group,and high-dose(800 mg/L) FSC group.MTT assay was used to observe cell proliferation rate.Immunohistochemistry was used to detect the positive signals of Ki67.The protein level of p-Akt,Akt,androgen receptor(AR) and CK18 was measured by using Western Blot assay.Results Compared with the control group,the proliferation rate of low-dose,mid-dose and high-dose groups significant manner increased(P<0.01).The positive signals of Ki67,the protein level of AR and p-Akt/Akt in FSC groups significantly increased(P<0.01) while the level of CK-18 significantly decreased(P<0.01) in dose-dependent manner(P<0.01 or P<0.05).Conclusion FSC may promote proliferation and differentiation of sertoli cells in young rats by regulating the level of AR,activating the PI3 K/Akt signaling pathway and increasing the phosphorylation of Akt.
