摘要 目的:研究低能量半导体激光生物刺激对雪旺细胞生物学行为的影响,筛选出雪旺细胞最适宜的激光治疗参数。方法:选用大鼠雪旺细胞(RSC96细胞系),给予低能量半导体激光生物刺激,建立低能量半导体激光雪旺细胞生物刺激模型。A组:空白对照组,即无激光照射;B组:250mW激光照射;C组:500mW激光照射;D组:1000mW照射。Annex in V/PI染色流式细胞仪测定半导体激光刺激对雪旺细胞调亡及细胞周期的影响。应用Western blot和ELISA检测低能量半导体激光刺激对雪旺细胞合成及分泌NGF及BDNF的影响。结果:低能量半导体激光在250mW参数照射下雪旺细胞的凋亡比例为0. 13%±0. 01%,细胞周期S期比例为7. 52%±1. 23%;NGF及BDNF蛋白表达量最高,较空白对照组分别增加1. 20倍和1. 18倍。250mW激光参数照射下雪旺细胞分泌NGF和BDNF的量分别为49ng/L和312 ng/L均高于对照组(A:31ng/L,B:49ng/L,C:35ng/L,D:25ng/L)。结论:在250mW低能量半导体激光生物刺激促进雪旺细胞增殖和抑制雪旺细胞调亡、细胞粘附及合成神经营养因子的能力。 Objective: To examine the putative regulatory effect of LLLT on the biological behaviors of Schwann cells, and examine the best suitable stimulation parameter of low level laser diode laser therapy. Method: The growth of Schwann cells was characterized using a flow cytometers (FACS). The protein levels of NGF and BDNF in Schwann cells were assayed by Western blotting and ELISA test. Results: Schwann cell under the irradiation Low level laser therapy with the parameter of 250mW proportion of apoptosis was 0.13%±0.01%;cell cycle S phase was 7.52%±1.23%. the secretion protein level of NGF and BDNF was 1.2 and 1.8 time higher than the control group. Conclusion: Low level laser therapy of 250mW can promote the proliferation, adhesion and apoptosis of Schwann cells, and promote the synthesis and secretion of NGF and BDNF protein level.
