TUSC3在乳腺癌中的表达及对乳腺癌细胞增殖能力的影响

Expression of TUSC3 in breast cancer and its effect on proliferation of breast cancer cells

目的探讨TUSC3基因在乳腺癌组织、乳腺癌细胞中的表达水平及对乳腺癌细胞增殖能力的影响。方法采用免疫组化方法检测乳腺癌组织和正常乳腺组织中TUSC3基因的表达;采用Western blot方法检测TUSC3在乳腺癌细胞中的表达;通过MTT实验分别观察转染后TUSC3表达高/低的乳腺癌细胞的增殖能力。结果 TUSC3在乳腺癌及乳腺癌旁组织中存在差异表达(χ2=5.926,P<0.05);与对照组比较,TUSC3表达高时,细胞增殖能力下降(P<0.05),TUSC3表达低时,细胞增殖能力上升(P<0.05)。结论 TUSC3基因在乳腺癌组织及乳腺癌旁组织中存在表达差异,并且上调TUSC3的表达可以抑制乳腺癌细胞的增殖能力,但其在乳腺癌发生发展中的作用及机制有待进一步研究。

Objective To investigate the expression level of TUSC3 gene in breast cancer tissues and cells and its effect on the proliferation of breast cancer cells.Methods The expression of TUSC3 gene in breast cancer and normal breast tissue was detected by immunohistochemical method, the expression of TUSC3 in breast cancer cells was detected by Western blot, and the proliferation ability of breast cancer cells with high or low TUSC3 expression was observed by MTT assay.Results TUSC3 was differentially expressed in breast cancer and adjacent tissues (χ^2=5.926, P <0.05);Compared with the control group, the proliferation ability of cells with higher TUSC3 expression was decreased ( P <0.05), and the proliferation ability of cells with lower TUSC3 expression was increased ( P <0.05).Conclusion The expression of TUSC3 gene was different in breast cancer tissues and adjacent tissues.Upregulation of TUSC3 expression could inhibit the proliferation of breast cancer cells, but its role and mechanism in the occurrence and development of breast cancer need further study.
