摘要
目的本研究旨在探讨linc-OIP5与乳腺癌恶性程度的关系,以及对乳腺癌细胞生物学功能的影响。方法采用RT-PCR方法检测linc-OIP5在不同恶性程度的乳腺癌细胞系中的表达情况;采用转染si RNA方法干扰乳腺癌细胞中的linc-OIP5基因,后以RT-PCR方法检测干扰效率;采用CCK8方法检测乳腺癌细胞的增殖能力;以及Transwell细胞侵袭实验检测细胞的侵袭能力。结果乳腺癌细胞中linc-OIP5的表达量与细胞的恶性程度呈正相关。在细胞功能实验中,与对照组相比,linc-OIP5干扰组细胞的增殖和侵袭能力均减弱。结论 Linc-OIP5在乳腺癌中起到致癌基因的作用,可以调控乳腺癌细胞的增殖和侵袭能力。
Objective To investigate the relationship between linc-OIP5 and the malignancy of breast cancer, and the effect of linc-OIP5 on the biological function of breast cancer cells. Methods RT-PCR was used to detect the expression of linc-OIP5 in breast cancer cell lines with different malignant degrees.The linc-OIP5 in breast cancer cells was knocked down by siRNA, and the interference efficiency was detected by RT-PCR.The proliferation activity of breast cancer cells was detected by CCK8 assay, and the invasion ability was measured by Transwell cell invasion assay.Results The relative expression of linc-OIP5 was positively correlated with the malignant degree of breast cancer cells.In the cell function experiment, the proliferation and invasiveness of the cells in the linc-OIP5 knockdown group were decreased compared with the control group. Conclusion Linc-OIP5 plays an oncogenic role in breast cancer and could regulate the proliferation and invasiveness of breast cancer cells.
