Expression and significance of CENPC and CENPT in breast cancer

目的本研究旨在分析着丝粒蛋白C(CENPC)和着丝粒蛋白T(CENPT)在乳腺癌中的表达情况,探讨CENPC和CENPT与乳腺癌患者生存率的关系。方法运用GEPIA数据库以及蛋白印记方法(Western blot)检测CENPC和CENPT在乳腺癌组织和癌旁正常组织中的表达。同时,通过生物信息学分析CENPC和CENPT的相关性。结果通过GEPIA数据库分析1085例乳腺癌样本和291例癌旁正常样本CENPT在乳腺癌中的表达水平低于在癌旁正常组织中的表达,对9对乳腺癌及癌旁正常组织进行Western blot分析,与数据库结果一致,CENPT在乳腺癌以及癌旁正常组织中的表达差异有统计学意义(P<0.05),CENPC与CENPT的表达正相关,但是CENPC与CENPT的表达对BRCA患者的总体生存率影响无统计学意义(P>0.05)。结论 CENPT在乳腺癌中的表达水平低于邻近正常组织中的表达水平,并且CENPT的低表达可能是不良的预后分子标志物,为乳腺癌的诊断以及预后提供新的个体化治疗靶点。

Objective To analyze the expression of centromere protein C and centromere protein T in breast cancer, and to investigate the relationship between the expression of CENPC and CENPT with the survival rate of breast cancer patients.Methods The expression of CENPC and CENPT in breast cancer tissues and adjacent normal tissues was detected by GEPIA database and Western blotting.The correlation between CENPC and CENPT was analyzed by bioinformatics.Results By GEPIA database analysis of 1085 cases of breast cancer sample and 291 cases of adjacent normal sample, the expression level of CENPC and CENPT in breast cancer were lower than those in adjacent normal tissues.The expression of CENPT in breast cancer and adjacent normal tissues was significantly different in 9 case of cancer and adjacent normal organizations by western blotting, which was consistent with database analysis ( P <0.05).The CENPC and CENPT expression level of BRCA patients has no relationship with overall survival rate ( P >0.05).Conclusion The CENPT expression levels were lower in breast cancer than those in adjacent normal tissue, CENPT low expression may be a poor prognosis of molecular markers for breast cancer diagnosis and prognosis, which could provide a new individualized therapeutic target.
