屠宰场猪肉中猪霍乱沙门氏杆菌的分离鉴定及16S rRNA的序列分析 被引量:2

摘要 为了了解屠宰场生猪肉被沙门氏杆菌污染的情况,试验对采集的20份生猪肉进行细菌分离培养、选择性培养、生化试验、革兰氏染色、沙门氏杆菌外膜蛋白C(Omp C)基因PCR检测等方法进行鉴定,采用MegAlign软件绘制分离菌16S rRNA遗传进化树。结果表明:从屠宰场生猪肉中分离到一株疑似沙门氏杆菌,命名为GZ-Q1株; GZ-Q1株为革兰氏阴性的短小杆菌;生化试验结果与沙门氏杆菌的生化特性基本相符; Omp C基因PCR检测可见约204 bp的目的条带; GZ-Q1株与猪霍乱菌株(CP007639. 1)的同源性最高,为99. 9%。说明从屠宰场生猪肉中分离的GZ-Q1株为猪霍乱沙门氏杆菌。 In order to understand the contamination of raw pork from slaughterhouses by Salmonella, 20 samples of raw pork were identified by bacterial isolation, selective culture, biochemical test, Gram staining, Salmonella outer membrane protein C(Omp C) PCR deteection, and 16S rRNA phylogenetic tree of isolated bacteria was drawn by MegAlign software. The results showed that a suspected Salmonella strain named GZ-Q1 was isolated from raw pork in slaughterhouses. GZ-Q1 strain was Gram-negative Bacillus pumilus. The results of biochemical tests were basically consistent with biochemical characteristics of Salmonella. Omp C PCR showed a target band of about 204 bp. GZ-Q1 strain had the highest homolog}-(99. 9%) with swine cholera strain (CP007639. 1). It indicates that the GZ-Q1 strain isolated from raw pork in slaughterhouses is Salmonella choleraesuis.
