摘要
为了研究枯草芽孢杆菌(WB600)对铅暴露鲫鱼肠道的影响,试验选取健康的鲫鱼192尾,随机分为8组,分别为对照Ⅰ组,枯草芽孢杆菌阳性对照Ⅱ组(1×109cfu/g),铅暴露Ⅲ组、Ⅴ组、Ⅶ组(0. 05,0. 5,1 mg/L),益生菌治疗Ⅳ组、Ⅵ组、Ⅷ组(0. 05 mg/L Pb+WB600、0. 5 mg/L Pb+WB600、1 mg/L Pb+WB600)。60 d后取肠道组织,通过常规方法制作石蜡切片,测量肠道绒毛高度、宽度及肌层厚度,从而评价该菌株对铅暴露鲫鱼肠道组织的影响。结果表明:与Ⅰ组相比,Ⅱ组近端肠和远端肠的绒毛高度、宽度、肌层厚度均显著增加(P <0. 05);与铅暴露组相比,益生菌治疗组近端肠及远端肠的绒毛高度、宽度、肌层厚度均不同水平增加;与Ⅲ组相比,Ⅳ组远端肠绒毛宽度显著增加(P<0. 05),其他均无显著差异(P>0. 05);与Ⅴ组相比,Ⅵ组近端肠绒毛宽度和远端肠绒毛高度、肌层厚度显著增加(P<0. 05),其他均无显著差异(P>0. 05);与Ⅶ组相比,Ⅷ组近端肠绒毛宽度和远端肠肌层厚度显著增加(P<0. 05),其他均无显著差异(P>0. 05)。说明枯草芽孢杆菌对不同铅浓度环境中鲫鱼的肠道结构有积极影响,对缓解铅导致的肠道损伤有一定的作用。
In order to investigate the effects of the Bacillus subtilise WB600 on the intestines of lead exposed crucian carp,192 healthy crucian carp were randomly divided into eight groups,respectively,which were contrast group Ⅰ,positive contrast group Ⅱ of Bacillus subtilis( 1 ×10~9 cfu/g),lead exposure group Ⅲ,Ⅴ and Ⅶ( 0. 05,0. 5,1 mg/L),probiotics treatment groups Ⅳ,Ⅵ and Ⅷ( 0. 05 mg/L Pb + WB600,0. 5 mg/L Pb+WB600,1 mg/L Pb+WB600). Sixty days later,intestine tissues were collected to test for making paraffin section by conventional method to measure the height,width and muscle thickness of intestinal villus,so as to evaluate the effects of the Bacillus subtilis WB600 on the intestines of lead exposed crucian carp. The results showed that compared with group Ⅰ,the height,width,muscle thickness of proximal and distal intestine villus of group Ⅱ were significantly increased( P<0. 05).Compared with lead exposure group,the height,width,muscle thickness of proximal and distal intestine villus of probiotics treatment groups were increased respectively. Compared with group Ⅲ,the width of distal intestinal villus of group Ⅳ were significantly increased( P<0. 05),the other had no significant difference( P> 0. 05).Compared with group Ⅴ,the width of the proximal intestinal villus and the height and muscle thickness of the distal intestinal villus of group Ⅵ were significantly increased( P<0. 05),the other had no significant difference( P> 0. 05). Compared with group Ⅶ,the width of the proximal intestinal villus and muscle thickness of the distal intestinal villus of group Ⅷ were significantly increased( P<0. 05),the other had no significant difference( P> 0. 05). Thus,the Bacillus subtilis WB600 has a positive effect on the intestinal structure of crucian carp in different lead concentrations,and plays a role in alleviating the intestinal damage caused by lead.
