梅毒患者外周血单个核细胞miR-1204的表达及其功能预测分析 被引量:3

摘要 目的探讨梅毒患者外周血单个核细胞miR-1204的表达及其潜在功能,为进一步研究miR-1204在梅毒螺旋体感染中生物学功能以及作用机制奠定基础。方法收集梅毒患者外周血83例、健康人12例,密度梯度离心法分离PBMCs,采用RT-qPCR检测miR-1204;采用Target Scan7.1和DIANA对miR-1204靶基因进行预测并取交集的基因集合,最后通过Consensus PathDB对基因集合进行GO功能及KEGG信号转导通路富集分析。结果一期梅毒患者PBMCs中miR-1204表达水平显著上调,高于二期梅毒患者(P<0.05)、血清治愈患者(P<0.05),差异具有统计学意义。miR-1204在现症梅毒组中表达水平高于血清学治愈组(P<0.05),差异具有统计学意义;GO功能富集分析显示miR-1204靶基因主要参与调节细胞发育、细胞分化、血管通透性、趋化性、抗原刺激的炎症反应等生物学过程。信号转导通路富集分析显示miR-1204 Top10信号转导通路为间隙连接、趋化因子信号通路、ErbB信号通路、GnRH信号通路、非洲锥虫病、松弛素信号通路、NK细胞介导的细胞毒作用、乙型肝炎、磷脂酶D信号通路、致病性大肠杆菌感染。结论 miR-1204可能成为一期梅毒/评估梅毒治疗效果潜在的分子标志物,参与早期梅毒感染的发生。 Objective To investigate the expression and potential function of miR-1204 in peripheral blood mononuclear cells(PBMCs) of syphilis patients. Methods Peripheral blood samples of 83 syphilis patients and 12 healthy controls were collected. PBMCs were isolated by density gradient centrifugation,miR-1204 was measured by RT-qPCR. Using Target Scan7.1 and DIANA to predict the target genes of miR-1204 and take the intersected gene sets,finally the GO functional enrichment and KEGG pathway enrichment were performed by ConsensusPathDB software. ResultsThe expression of miR-1204 in PBMCs of primary syphilis patients was significantly up-regulated,higher than secondary syphilis patients (P<0.05) and serological cure patients (P<0.05). The expression of miR-1204 in the current syphilis group was higher than serological cure group (P<0.05). GO functional enrichment analysis showed that miR-1204 target genes were mainly involved in biological processes,such as the regulation of cell development, differentiation,vascular permeability,chemotaxis and inflammatory response to antigenic stimulus. Enrichment analysis of the signal transduction pathways revealed that the miR-1204 Top10 signal transduction pathways were gap junction,chemokine signaling pathway,ErbB signaling pathway,GnRH signaling pathway,African trypanosomiasis,Relaxin signaling pathway,Natural killer cell mediated cytotoxicity,Hepatitis B,Phospholipase D signaling pathway,and Pathogenic Escherichia coli infection. ConclusionmiR-1204 may be a potential biomarker for primary syphilis to evaluate the therapeutic effection,and may be involved in the early syphilis infection.
