摘要
为研究鳞片不同部位以及不同植物激素种类与浓度对风信子鳞片分化不定芽的影响,本研究以‘Blue Jacket’、‘Wood Stock’、‘Fondant’和‘Aiolos’4个风信子品种为研究对象,用其鳞片上、下两部分进行组织培养,培养基中分别添加不同浓度的6-BA (0.2 mol/L, 0.5 mol/L, 1.0 mol/L, 1.5 mol/L, 2.0 mol/L)、KT (0.2 mol/L, 0.5 mol/L, 1.0 mol/L, 1.5 mol/L, 2.0 mol/L)、TDZ (0.01 mol/L, 0.05 mol/L, 0.1 mol/L, 0.2 mol/L,0.4 mol/L)和ZT (0.2 mol/L, 0.5 mol/L, 1.0 mol/L, 1.5 mol/L, 2.0 mol/L)。结果表明:风信子鳞片不同部位对其分化不定芽的影响不同,但下部鳞片更适宜作为外植体来诱导分化不定芽;不同植物激素的种类与浓度对风信子鳞片分化不定芽的影响也不同,其中,诱导效果最好的种类是TDZ和ZT。同时,得到4种风信子的最适培养基MS+ZT 1.0 mg/L+IBA 0.1 mg/L (‘Blue Jacket’和‘Fondant’),MS+0.1 mg/L TDZ+0.1 mg/L IBA(‘Wood Stock’),MS+0.5 mg/L ZT+0.1 mg/L IBA (‘Aiolos’)。本研究对风信子组织培养体系的优化方面提供了科学依据。
To study influenc e of different dosage of plant hormones and different parts of scales to hyacinth adventitious buds differentiation, we chosed four types for tissue culture as subject investigated in this study,respectively ’Blue Jacket’,’Wood Stock’,’Fondant’, and ’Aiolos’. The upper and lower parts of the scale were cultured in different concentrations of 6-BA(0.2 mol/L, 0.5 mol/L, 1.0 mol/L, 1.5 mol/L, 2.0 mol/L), KT(0.2 mol/L,0.5 mol/L, 1.0 mol/L, 1.5 mol/L, 2.0 mol/L), TDZ(0.01 mol/L, 0.05 mol/L, 0.1 mol/L, 0.2 mol/L, 0.4 mol/L) and ZT(0.2 mol/L, 0.5 mol/L, 1.0 mol/L, 1.5 mol/L, 2.0 mol/L). The results showed that the different parts of the hyacinth scales had different effects on the differentiated adventitious buds, but the lower parts of scales were more suitable as explants to induce the differentiation of adventitious buds. The results also indicated that the different plant hormones types and densities on the differentiation of adventitious buds of hyacinth cultivars were different, and the best types of induction among them were TDZ and ZT. Meanwhile, the optimum medium for 4 hyacinth species was obtained: MS+1 mg/L ZT+0.1 mg/L IBA(’Blue Jacket’ and ’Fondant’), MS+0.1 mg/L TDZ+0.1 mg/L IBA(’Wood Stock’), and MS+0.5 mg/L ZT+0.1 mg/L IBA(’Aiolos’). This study might provide a scientific basis for the optimization of hyacinth tissue culture system.
