摘要
目的探讨茯苓与生僵蚕、麸炒僵蚕的水提醇沉粗提物对紫外线(UVB)诱导豚鼠皮肤色素沉着的干预作用。方法选定茯苓与生僵蚕、麸炒僵蚕3种中药,经过水提醇沉后制成乳膏,外用于花色豚鼠UVB诱导色素沉着模型,采用肉眼观察并积分,评价豚鼠皮肤反应,采用苏木精-伊红(HE)染色观察外用上述中药后黑素颗粒数量变化,并对豚鼠皮肤组织中酪氨酸酶(TYR)的含量进行检测。结果与模型组比较,茯苓组、生僵蚕组和茯苓配生僵蚕组的豚鼠皮肤色素明显变淡,组织黑素颗粒明显减少,组织匀浆液中TYR含量明显降低(P <0.05)。结论茯苓、生僵蚕和茯苓配生僵蚕具有抑制色素沉着的作用,其中生僵蚕抑制效果最佳,作用机制可能与其能够抑制TYR的分泌有关。
Objective To explore the interventional effects of the extract made by ethanol sedimentation of Poria cocos,Bombyx batryticatus and Bombyx batryticatus fried with bran on guinea pig pigmentation models induced by UVB. Methods The cream prepared with Poria cocos,Bombyx batryticatus and Bombyx batryticatus fried with bran after ethanol sedimentation was externally used on guinea pig pigmentation models induced by UVB. Evaluation of the skin reaction of guinea pigs was carried out by grading based on gross observation results. Quantitative changes of melanin granules were observed using HE staining. The content of TYR in guinea pigs’ skin tissue was tested.Results Compared with the model group, the skin pigmentation of guinea pigs of the Poria cocos, Bombyx batryticatus and Poria cocos combined with Bombyx batryticatus groups were noticeably lighter, the number of melanin granules and the contents of TYR of the three groups distinctly decreased(P < 0.05). Conclusion Poria cocos, Bombyx batryticatus and Poria cocos combined with Bombyx batryticatus have the effect of inhibiting hyperpigmentation. The inhibitory effect of Bombyx batryticatus is the best among the three groups, and its mechanism may be related to inhibition of the secretion of TYR.
