摘要
分析米象代谢解毒相关基因谷胱甘肽硫转移酶(GST)的表达,获得米象基因遗传信息,进而从本质上全面揭示米象的磷化氢抗性。米象敏感品系经磷化氢不同浓度熏蒸和不同时间熏蒸处理,2个基因经LC_(30)浓度磷化氢熏蒸的相对表达量显著高于其他2个浓度;4个GSTs基因经LC_(50)浓度磷化氢熏蒸的相对表达量显著高于对照组和LC_(30)、LC_(10)处理组;3个基因相对表达量均显著低于对照组。7个GSTs基因随着熏蒸时间的增加,相对表达量逐渐升高。米象GSTs基因在不同发育阶段的相对表达模式中,SoGSTd1基因在四龄幼虫阶段的相对表达量最高;SoGSTe1基因在成虫阶段的相对表达量最高;SoGSTe2和SoGSTe5基因在四龄幼虫阶段的相对表达量最大,SoGSTe6基的相对表达量在三龄幼虫阶段最大。米象13个GSTs基因在不同组织部位的相对表达模式中。米象13个GSTs基因中的11个在中肠、脂肪体或马氏管中的相对表达量均显著高于头部和表皮中的表达量。
Through transcriptome technology,several resistant genes related to S.oryzae such as glutathione S-transferase,cytochrome were analyzed and the transcriptome genetic information was obtained,revealing the resistance of insectsin nature at transcriptome level is gaining a huge interest among the researchers.The S.oryzae sensitive strains of different concentrations of phosphine fumigation and fumigation treatment at different time were investiagted.The concentration of two genes was significantly higher than the other two concentrations by the concentration of LC 30,and the relative expression of the four GS tsgenes was significantly higher than that of the control group and the LC 30,LC10 treatment group,and the relative expression of the three genes was significantly lower than that of the control group.The relative expression level of 7 Gstsgenes increased gradually with the increase of fumigation time.The relative expression of the SoGSTd1 in the fourinstar stage was the highest in the relative expression pattern of the GS tsgene in the different developmental stages,and the relative expression of the SoGSTe1 gene in the adult stage was the highest.The relative expression of the SoGSTe2 and SoGSTe5 genes in the four instar stage was the largest,and the relative expression of the SoGSTe6 gene in the three instar larvae was maximum.13 Gstsgenes of S.oryzae were differently expression in different tissues.The relative expression level of13 Gstsgenes in midgut,fat body or malpighian tubules in S.oryzae was significantly higher than that in the head and cuticle.The relative expression level of11 genes in the midgut,fat body or malpighian tubules was higher than that in the head and cuticle.
