摘要
目的:应用Box-Behnken效应面法优化陕西法制天南星的提取工艺,并研究其抗肿瘤活性。方法:在单因素实验基础上,以总多糖和总黄酮的总评归一值为评价指标,料液比(X1)、浸提时间(X2)和提取温度(X3)为考察对象,利用星点设计效应面法优化陕西法制天南星提取工艺,同时观察提取物对体外抗肿瘤作用。结果:优化后的最佳工艺为料液比1∶25.0,提取时间144 min,提取温度89.5℃,陕西法制天南星水提物对A549的抑制率IC50为1.126mg/mL,《中国药典》方法制天南星水提物对A549的抑制率IC50为1.925 mg/mL。结论:Box-Behnken效应面法优化陕西法制天南星提取工艺方法可行,制天南星水提物能抑制体外培养肺癌A549细胞的生长。
Objective: The Box-Behnken response surface method was used to optimize the extraction pro-cess of Arisaematis rhizoma preparatum (in Shaanxi) and its anti-tumor activity was studied. Methods: Basis on single factor experiment, the extraction rate of polysaccharide and flavonoids as the evaluation index, the ratio of material to liquid (XI), extraction time (X2) and extraction temperature (X3) as the research object, the optimization of extraction process of Arisaematis rhizoma preparatum (in Shaanxi) was used to observe the effect of central composite design, inhibition rate of A549 was used to evalution anti-tumor effect in vitro. Results: The best op-timized process for solid-liquid ratio 1:25.0, extraction time 144 min, extraction temperature 89.5℃, the IC50 of Arisaematis rhizoma preparatum (in Shaanxi) extract obviously inhibited A549 in vitro at 1.126 mg/mL, and the IC50 of Arisaematis rhizoma preparatum (ChP) extract obviously inhibited A549 in vitro at 1.925 mg/mL. Conclu-sion: Box-Behnken effect surface method is feasible to optimize the extraction process of Arisaematis rhizoma preparatum, and water extract can inhibit the growth of A549 cells in vitro.
