Optimization of the Processing Parameters for the Extraction of the Decumbent Corydalis Tuber Formula using Box-Behnken Experimental Design

摘要 目的:为有效控制制剂的质量,通过建立总生物碱的含量测定方法,优化夏天无配方颗粒的水提取工艺参数。方法:通过单因素实验筛选,应用Box-Behnken响应面法,对粉碎程度、加水量、煎煮时间、煎煮次数4个因素进行多元线性回归,建立回归模型,分析各因素对出膏率、夏天无总生物碱含量转移率两个指标的影响。结果:粉碎程度和加水量是夏天无配方颗粒提取工艺的关键因素,其次是煎煮次数和煎煮时间。夏天无配方颗粒的最佳提取工艺为:夏天无药材粉碎(过8mm筛网),加水提取3次,每次提取60min,每次加入10倍量水。结论:经Box-Behnken设计法建立的数学模型的预测值与实验观察值基本相符,优选的提取工艺参数可应用于夏天无配方颗粒的提取环节。 Objective:To preference the extraction process of the Decumbent Corydalis Tuber formula,establish the determination of the content of total alkaloids,provide a reference for the effective control of the quality of the preparation.Methods:Based on the results of single-factor-at-a-time-technique,Box-Behnken experimental design combined with response surface methodology was employed,with the degree of grinding(A),the water addition(B),the eatraction times(C),the eatraction time(D)as the independent variables.The response variable is the extraction ratio and the con tent transfer rate of total alkaloids.Results:It is pivotal factor with degree of grin ding and the water additi on,secondly,eatraction times and the eatraction time,the optimization processing parameters is as follows:the decumbent corydalis tuber formula was smashed to 8 mm,extract 3 times using pure water,each for 60 min.Conclusion:The observed responses are in close agreement with the predicted values of the mathematic models,and the Box-Behnken experimental design is suitable for optimizing the extraction of the Decumbent Corydalis Tuber formula.
