摘要 目的:利用ITS序列对不同来源辽细辛进行DNA分子鉴定。方法:提取辽细辛植物的DNA,对其ITS序列进行逆转录-聚合酶链式反应(RT-PCR)扩增与测序,且从GenBank中下载上述所属植物的ITS序列,进行辽细辛的比对。结果:从NJ系统发育树分析看出,两属4个物种的26个样本被聚为3大支。细辛植物自成1支,汉城细辛属1支,马兜铃属植物1支。结论:表明ITS序列可以对细辛原植物进行鉴别,且在近缘物种鉴定方面具有较大应用价值。 Objective:To characterize DNA molecules from different sources of Asarum by ITS sequences.Methods:The DNA of the plant was extracted and its ITS sequence was amplified and sequenced by reverse transcription-polymerase chain reaction(RTPCR).The ITS sequences of the above-mentioned plants were downloaded from GenBank.Comparison.Results:From the analysis of the NJ phylogenetic tree,26 samples of 2 genera and 4 species were clustered into three major branches.Asarum plants from one,Seoul Asarum is a branch.Aristolochia plant 1.Conclusion:It is shown that the ITS sequence can be used to identify the original plant,and it has great application value in the identification of the similar species.
