摘要 目的研究TRPC、ORAI1和NCX三种钙调控蛋白在血管平滑肌细胞、内皮细胞中表达的差异。方法通过实时荧光RT-PCR和Western blot分别检测了TRPC (TRPC1、TRPC3、TRPC4、TRPC5和TRPC6)、ORAI1和NCX (NCX1和NCX2)钙调控蛋白在血管平滑肌细胞与血管内皮细胞中的m RNA和蛋白表达水平。结果TRPC6、ORAI1在血管内皮细胞中m RNA和蛋白表达水平均高于血管平滑肌细胞;TRPC4、TRPC5和NCX2在血管平滑肌细胞中的m RNA和蛋白表达水平均高于在血管内皮细胞,尤其是TRPC4、TRPC5在蛋白表达水平差异有统计学意义(P <0.01)。结论通过差异性表达分析筛选出内皮细胞中高表达的TRPC6、ORAI1和平滑肌细胞中高表达的TRPC4、TRPC5,可能通过选择性钙通道蛋白的调控,从而抑制平滑肌细胞增殖和促进再内皮化,为动脉粥样硬化的预防和治疗、血管支架植入后再狭窄的发生发展提供理论依据。 Objective To investigate the differences in expression of three calcium regulatory proteins, TRPC, ORAI1 and NCX, in vascular smooth muscle cells and endothelial cells. Methods mRNA and protein expression of TRPC (TRPC1, TRPC3, TRPC4, TRPC5 and TRPC6), ORAI1 and NCX (NCX1 and NCX2) calcium regulatory proteins in vascular smooth muscle cells and vascular endothelial cells were detected by real-time fluorescent RT-PCR and Western blot, respectively.Results The mRNA and protein expression levels of TRPC6 and ORAI1 in vascular endothelial cells were higher than those in vascular smooth muscle cells;the mRNA and protein expression levels of TRPC4, TRPC5 and NCX2 in vascular smooth muscle cells were higher than those in vascular endothelial cells,and TRPC4, TRPC5 had significant differences.have significant differences at protein expression level(P<0.01) Conclusions Highly expressed TRPC6, ORAI1 in endothelial cells and highly expressed TRPC4 , TRPC5 in smooth muscle cells were screened by differential expression analysis, which may inhibit the proliferation of smooth muscle cells and promote re-endothelialization through the regulation of selective calcium channel proteins.We provide a theoretical basis for the prevention and treatment of atherosclerosis and the occurrence and development of restenosis after vascular stent implantation.
