香茅精油对D-半乳糖联合三氯化铝所致老年痴呆小鼠神经细胞损伤的影响及其机制研究 被引量:4

Effects and mechanism of Citronella oil on neurocyte injury in senile dementia mice induced by D-galactose combined with aluminum trichloride

摘要 目的观察香茅精油对D-半乳糖联合三氯化铝(Al Cl_3)所致老年痴呆(SDT)小鼠神经细胞损伤与炎症因子表达的影响及可能机制。方法昆明小鼠皮下注射5%D-半乳糖120 mg·kg^(-1)加灌胃Al Cl3200 mg·kg^(-1)共10周制备SDT小鼠;另选15只正常小鼠作为正常组。按照体重将造模成功小鼠随机分为5组:模型组、对照组和低、中、高3个剂量实验组,每组15只。正常组与模型组灌胃0. 9%Na Cl,对照组灌胃脑复康0. 5 g·kg^(-1),低、中、高3个剂量实验组灌胃0. 4,0. 6和0. 8 m L·kg^(-1)香茅精油,灌胃体积均为10μL·g^(-1),从造模第11周开始给药,每日1次,连续给药4周。以酶联免疫吸附法测定脑组织肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)和白细胞介素-1β(IL-1β)的含量。结果正常组、模型组、对照组和低、中、高3个剂量实验组的TNF-α含量分别为(46. 89±8. 26),(74. 87±9. 77),(59. 32±8. 73),(63. 48±7. 22),(57. 47±6. 82)和(58. 23±7. 97) pg·m L^(-1);这6组的IL-6含量分别为(56. 16±5. 91),(85. 45±8. 53),(68. 46±5. 67),(68. 55±6. 39),(64. 23±5. 96)和(66. 04±5. 43) pg·m L^(-1);这6组的IL-1β含量分别为(44. 96±4. 81),(57. 10±4. 91),(51. 74±5. 92),(57. 39±3. 79),(51. 32±4. 13)和(50. 51±5. 17) pg·m L^(-1)。模型组与正常组比较,上述指标的差异均有统计学意义(均P <0. 01);对照组和3个剂量实验组与模型组比较,除低剂量实验组(IL-1β)外,上述指标的差异均有统计学意义(P <0. 05或0. 01)。结论香茅精油可改善D-半乳糖联合Al Cl3所致SDT小鼠神经细胞损伤,降低脑组织炎症反应。 Objective To investigate the effect of Citronella oil on damage of brain cells and inflammatory factor expression of brain tissue in senile dementia( SDT) mice induced by D-galactose and aluminum trichloride( Al Cl3) and explore its mechanism.Methods The KM mice with half males and half females were intraperitoneal injection of D-galactose and oral aluminum trichloride( Al Cl3) to make disease models for consecutive 10 weeks;15 of normal mice were as the normal group.The model mice were randomly divided into five groups: model group,control group( 0.5 g·kg^-1 piracetam) and low,middle,high doses( 0.4,0.6,0.8 m L·kg^-1 Citronella oil) experimental groups,each group had 15 mice.The volume of intragastric administration was 10 μL · g^-1.Drugs were administered from the 11 th week of modeling.Continuous administration once a day for 4 weeks.The level of tumor necrosis factor-α( TNF-α),interleukin-6( IL-6),interleukin-1β( IL-1β) in brain tissue homogenate were determined by enzyme-linked immuno sorbent assay.Results The level of TNF-α in the normal group,model group,control group and low,middle,high doses experimental groups were respectively( 46.89±8.26),( 74.87±9.77),( 59.32±8.73),( 63.48±7.22),( 57.47±6.82),( 58.23±7.97) pg·m L^-1;the level of the IL-6 in the 6 groups were respectively( 56.16±5.91),( 85.45±8.53),( 68.46±5.67),( 68.55±6.39),( 64.23±5.96),( 66.04±5.43) pg·m L^-1;the level of IL-1β in the 6 groups were respectively( 44.96±4.81),( 57.10±4.91),( 51.74±5.92),( 57.39±3.79),( 51.32±4.13),( 50.51±5.17) pg·m L^-1.Compared between the normal group with model group,the difference of the factors were significant( all P < 0.01).Compared between control group and low,middle,high doses experimental groups with model group,the difference of the factors( except for IL-1β in the low experimental group) were significant( P < 0.05,P < 0.01).Conclusion Citronella oil improve the nerve cell injury of senile dementia mice of Alzheimer’s type induced by D-galactose and Al Cl3,alleviate the infla-mmation response in brain tissue.
