昆布多糖对D-半乳糖诱导的衰老小鼠肝损伤的保护作用及其分子机制研究 被引量:1

Protective effects of laminarin on liver injury induced by D-galactose in aging mice and its molecular mechanism

摘要 目的:研究昆布多糖对D-半乳糖(D-galactose, D-Gal)诱导小鼠衰老的保护作用和分子机制。方法 :选取体质量(20±2) g的SPF级昆明小鼠100只,雌雄各半,随机分为5组(每组20只),即对照组(Control组)、衰老模型组(D-Gal组)、葛根素阳性对照组(Positive组)、昆布多糖高剂量组(Lam-H组)和昆布多糖低剂量组(Lam-L组)。造模给药结束后,通过水迷宫实验检测小鼠的记忆行为; HE染色观察小鼠脑、肝组织的病理变化;最后检测小鼠脑、肝组织中P16、P21蛋白和mRNA水平。结果:Lam-H组、Lam-L组均缩短了D-Gal诱导的衰老小鼠到达水迷宫平台的时间,减轻了小鼠脑、肝组织病理学损伤,并下调脑、肝组织中P16、 P21蛋白和mRNA含量。结论:昆布多糖能够改善D-Gal诱导衰老小鼠的记忆行为,并保护D-Gal诱导衰老小鼠脑、肝组织学形态,其机制可能与调节脑、肝中P16、P21蛋白和mRNA表达水平有关。 Objective : The protective effect and molecular mechanism of laminarin on D-galactose(D-Gal)-induced aging in mice were studied. Methods : A total of 100 SPF Kunming mice of (20±2) g, male and female, were randomly divided into 5 groups(20 in each group), namely the control group(Control group) and the aging model group(D-Gal group), puerarin positive control group(Positive group), laminarin high dose group(Lam-H group) and laminarin low dose group(Lam-L group). After the administration of the model, the memory behavior of the mice was detected by water maze test. The pathological changes of the brain and liver tissues of the mice were observed by HE staining. Finally, the levels of P16 and P21 protein and mRNA in the brain and liver tissues of the mice were detected. Results : Both Lam-H and Lam-L groups shortened the time that D-Galinduced aging mice reached the water maze platform, reduced the pathological damage of brain and liver tissues in mice, and down-regulated P16, P21 protein and mRNA content in brain and liver tissues. Conclusion : Laminarin can improve the memory behavior and protect the brain and liver histology in senescence mice induced by D-galactose, and the potential mechanism may be associated with the regulation of P16 and P21 expresions.
