摘要 以日本独头白菊‘精诚’植株的顶芽、带腋芽茎段为外植体进行离体快繁研究。结果表明:以0.1%Hg Cl2为表面消毒剂,最适消毒时间为5 min,初代培养中,顶芽的培养效果比带腋芽茎段更好;以顶芽为外植体,初代培养的最适培养基为MS+0.5 mg/L 6-BA+0.1 mg/L NAA,其平均有效芽诱导数为5.87;在增殖培养中,以带腋芽茎段诱导丛生芽,其最适增殖培养基为MS+1.0 mg/L 6-BA+0.3 mg/L NAA,增殖系数为6.92;以叶片切段诱导不定芽,其最适增殖培养基为:MS+1.0 mg/L 6-BA+0.3 mg/L NAA,增殖系数为3.59;以叶柄诱导不定芽,其最适增殖培养基为:MS+1.5 mg/L 6-BA+0.15 mg/L NAA,增殖系数为0.49;最适生根培养基为:1/2MS+0.35 mg/L IBA,生根率100%。本研究结果可以为日本独头白菊‘精诚’的工厂化生产提供技术支持,具有重要的实践价值。 In vitro rapid propagation of Japanese single-headed white chrysanthemum ‘Jingcheng’ was made by using apical buds and stem segments with axillary buds as explants. In primary culture, explants were best sterilized with 0.1% mercuric chloride as surface disinfectant for 5 minutes, and the apical buds had better culture results than the axillary buds. The optimal medium for culture of the apical buds was MS+ 0.5 mg/L 6-BA and 0.1 mg/L NAA, which induced 5.87 effective shoots by average. In proliferation culture, the shoots from the primary culture were cut into stem segments with axillary buds and induced to generate clustered shoots on the optimal medium of MS+1.0 mg/L of 6-BA+0.3 mg/L of NAA, and its multiplication coefficient was 6.92. The optimal medium for inducing adventitious shoots from the leaf was MS+1.0 mg/L of 6-BA+0.3 mg/L of NAA, and the multiplication coefficient of 3.59. The optimal proliferation medium for inducing adventitious shoots from the petioles of the leaves was MS+1.5 mg/L 6-BA+0.15 mg/L NAA, and the multiplication coefficient was only 0.49. The optimum medium for root induction was 1/2 MS+0.35 mg/L IBA and the root inducing rate was upto 100%. These results will provide technical support for commercial production of elite seedlings of Japanese single-headed white chrysanthemum ‘Jingcheng’, and has important practical value.
