乳腺癌中DEAH盒解旋酶16的表达及临床意义 被引量:1

Analysis of DEAH-Box Helicase 16 Expression and Its Clinical Significance in Breast Cancer

摘要 目的揭示乳腺癌组织中DEAH盒解旋酶16 (DHX16)基因表达变化及其与乳腺癌患者预后的关系。方法利用c BioPortal分析DHX16基因突变、拷贝数变化、mRNA表达变化、m RNA差异表达与预后的关系,DHX16基因甲基化水平与其mRNA水平的关系。利用UALCAN数据平台分析乳腺癌组织与正常乳腺组织中DHX16基因的差异表达。利用LinkedOmics分析乳腺癌组织中DHX16表达与临床指标的相关性,DHX16基因拷贝数变化与mRNA水平的关系。结果 cBioPortal结果显示,16%(170/1 093)的乳腺癌患者发生了DHX16基因改变,包括基因突变、拷贝数变化、mRNA水平改变,其中DHX16 mRNA水平上调(超过对照组平均值2倍)的患者比例为12%,且DHX16高表达不利于患者预后(P <0.01);DHX16 mRNA表达水平与其基因甲基化水平呈负相关(r=-0.32,P <0.05)。UALCAN结果显示,乳腺癌组织中DHX16基因表达水平显著高于正常乳腺组织(P <0.001)。Linked Omics结果显示,乳腺癌患者中DHX16表达水平分别受PAM50分子分型、种族、病理分期、T分期和M分期影响(均P <0.05);DHX16 mRNA表达水平与其基因拷贝数变化呈正相关(r=0.56,P <0.01)。结论乳腺癌中DHX16基因拷贝数增加和低甲基化促进其高表达,DHX16高表达不利于预后,可作为乳腺癌预后的潜在标志物。 Objective To uncover the expression pattern of DEAH-box helicase 16(DHX16)in breast cancer and to evaluate its relationship with prognosis of breast cancer patients.Methods CBioPortal was used to analyze DHX16 gene mutation,copy number variation,mRNA expression and its relationship with prognosis,and the correlation between methylation levels and mRNA expression.UALCAN was used to determine the differential expression of DHX16 in breast cancer and healthy tissues.LinkedOmics was used to determine the correlation between DHX16 expression and various indicators of breast cancer.Results Results from cBioPortal indicated that DHX16 gene showed mutations and copy number changes in 16%patients,and an increase in DHX16 expression in 12%patients.High level of DHX16 mRNA was correlated with poorer prognosis of patients(P<0.01).Results from UALCAN showed that DHX16 mRNA levels were significantly higher in breast cancer tissue than those in normal tissue(P<0.001).Results from LinkedOmics showed that DHX16 mRNA levels were significantly affected by PAM50 typing,race,pathology,TNM stage,distant metastasis stage,and infiltration stage(P<0.05 for all).DHX16 mRNA levels were positively correlated with gene-copy number alteration(r=0.56,P<0.01),but negatively correlated with DNA methylation(r=-0.32,P<0.05).Conclusion Increased DHX16 copy number and hypomethylation promote its expression,and high expression of DHX16 is not conducive to a better prognosis in breast cancer patients.Therefore,DHX16 can be used as a prognostic marker.
