TiO_2光动力疗法治疗鼻咽癌移植瘤模型的初步研究

摘要 目的:观察TiO_2光动力疗法对鼻咽癌移植瘤的治疗作用,研究其对肿瘤的抑制作用。方法:建立鼻咽癌移植瘤模型,随机分组,观察注入纳米TiO_2悬液的移植瘤组织在330~400 nm波长紫外光辐照下的组织病理学与超微结构的变化及肿瘤细胞凋亡情况。结果:光镜和透射电镜下观察,光动力治疗后治疗组移植瘤组织的局部炎性反应、坏死灶形成及瘤细胞凋亡情况均显著高于对照组,差异有统计学意义(P<0.01),程度随TiO_2浓度递增而增强。结论:TiO_2光动力疗法通过介导炎性反应和诱导细胞凋亡发挥抑癌效果,这可能也是其治疗鼻咽癌的重要机制之一,移植瘤模型实验提示其是治疗鼻咽癌的可行手段。 Objective To explore the inhibiting effect of photocatalytic nano-TiO2 on nasopharyngeal carcinoma xenograft in nude mice and underlying mechanism.Method Nude micc bearing human nasopharyngeal carcinoma xenograft were randomly assigned.The nano-TiO2 suspension was injected into xenografts,and after UV light with the wave length of 30-440 nm.The histopathological and ultrastructural changes of the transplanted tumor and the apoptosis of the tumor cells were observed.Results Light microscopy and transmission electron microscopy showed that the local inflammatory response,necrosis and apoptosis of transplanted tumor tissues in the experimental group were significantly higher than those in the control group(P<0.01),and the degree increased with the increase of TiO2 concentration.Conclusion Photocatalytic nano-TiO2 played an important role in the inhibition of cancer by mediating inflammatory reaction and inducing cell apoptosis.This might be one of the important mechanisms for its treatment of nasopharyngeal carcinoma.The model experiment of transplanted tumor suggests that it was a feasible method for the treatment of nasopharyngeal carcinoma.
