银杏黄酮对胃癌模型大鼠VEGF、Bcl-2、Bax表达水平的影响 被引量:10

摘要 目的探究银杏黄酮对胃癌模型大鼠血管内皮生长因子(VEGF)、B淋巴细胞瘤-2基因(Bcl-2)、Bax蛋白表达水平的影响。方法选取30只SD健康雄性大鼠,20只大鼠进行胃癌模型的建立,建模成功18只,分为胃癌组和银杏黄酮组各9只,另外10只为正常组,对银杏黄酮组大鼠使用银杏黄酮进行干预,于大鼠腹腔注射100 mg/kg的银杏黄酮溶液10 m L/kg,1次/d,连续注射7 d;正常组和胃癌组大鼠使用生理盐水进行干预,剂量、频次与银杏黄酮组保持一致。Western blot法检测Bcl-2、Bax、表皮生长因子受体(EGFR)、AKT、哺乳动物雷帕霉素靶蛋白(m TOR)水平,酶联免疫吸附试验法检测VEGF、胃动素、胃泌素水平。结果 VEGF、Bcl-2水平:胃癌组>银杏黄酮组>正常组(P <0. 05); Bax水平:胃癌组<银杏黄酮组<正常组(P <0. 05);胃动素、胃泌素水平:胃癌组<银杏黄酮组<正常组(P <0. 05); EGFR/AKT/m TOR通路相关蛋白表达水平:胃癌组>银杏黄酮组>正常组(P <0. 05)。结论使用银杏黄酮对胃癌模型大鼠进行干预,能够明显调控VEGF、Bcl-2、Bax表达水平,为胃癌的临床治疗提供一定的理论帮助。 Objective To investigate the effects of Ginkgo Flavone on the expression of vascular endothelial growth factor(VEGF), B lymphoma-2 gene(Bcl-2) and Bax protein in gastric cancer model rats. Methods Thirty healthy male SD rats and 20 rats were selected to establish gastric cancer model. Eighteen rats were successfully established. They were divided into gastric cancer group and ginkgo flavone group, 9 rats in each group, and 10 rats in the other group were normal. Ginkgo flavone group was intervened with Ginkgo flavone. Ginkgo flavone solution of 100 mg/kg was injected into the abdominal cavity of rats 10 ml/kg once a day for 7 days. Normal and gastric cancer groups were injected with physiological salts. The dose and frequency of water intervention were consistent with that of Ginkgo flavone group. The levels of Bcl-2, Bax, epidermal growth factor receptor(EGFR), AKT and mammalian rapamycin target protein(m TOR) were detected by Western blot, and the levels of VEGF, motilin and gastrin were detected by ELISA. Results Levels of vascular endothelial growth factor and Bcl-2: gastric cancer group > ginkgo flavone group >normal group(P < 0. 05);Bax level: gastric cancer group < ginkgo flavone group < normal group(P < 0. 05);motilin and gastrin level: gastric cancer group < ginkgo flavone group < normal group(P < 0. 05);expression level of EGFR/AKT/m TOR pathway related protein: gastric cancer group > ginkgo flavone group > normal group(P < 0. 05). Conclusion The intervention of ginkgo flavone in gastric cancer model rats can significantly regulate the expression levels of VEGF, bcl-2 and Bax, providing theoretical help for the clinical treatment of gastric cancer.
