大鼠不同胚层来源成骨细胞生物学行为比较研究 被引量:1

摘要 目的:探讨大鼠不同胚层来源成骨细胞生物学行为的差异。方法:体外培养大鼠颌骨与股骨成骨细胞(MOBs与FOBs),通过MTT比色法检测细胞的增殖能力,采用碱性磷酸酶染色与活性测定、茜素红染色与定量比较细胞的成骨能力。应用荧光定量PCR及ELISA检测细胞成骨与成血管相关细胞因子的表达情况。结果:与FOBs相比, MOBs的增殖能力、成血管相关细胞因子Angpt-1和FGF-2的表达更强,但碱性磷酸酶的表达、体外矿化结节形成能力以及成骨相关基因ALP、RUNX2、Col-Iα1和OC的表达均较低。结论:MOBs与FOBs相比较,增殖力较强,分化能力较弱。 Objective:To study the difference of the biological behaviors of rat osteoblasts from distinct embryonic germ layers.Methods:Osteoblasts were isolated and cultured from mandible(MOBs)and femur(FOBs)of rats respectively.The proliferation of the cells was assessed by MTT assay.The property of osteogenic differentiation was evaluated by ALP staining and,Alizarin red staining and quantification analysis.The expression of osteogenesis-related genes and angiogenesis-related cytokines of the cells was evaluated by Real-time PCR and ELISA respectively.Results:Compared FOBs,MOBs showed higher proliferation ability,up-regulated the expression of angiogenesis-related cytokines Angpt-1 and FGF-2,but lower ALP activity and lower formation of mineralized nodules and lower expression of osteogenesis-related genes ALP,RUNX2,Col-Iαand OC.Conclusion:MOBs may have stronger and lower proliferation ability,lower osteogenic differentiation and pro-angiogenic property than FOBs.
