摘要
目的:利用生物信息学方法识别口腔扁平苔藓(OLP)发病过程中的生物学通路及关键基因或分子。方法:从GEO数据库中下载GSE38616,通过t检验结合Benjamini-Hochberg校正的方法筛选差异表达基因,并根据差异表达基因构建蛋白质互作网络,利用Cytoscape进行网络中心(Hub)节点识别,最后对差异表达基因进行富集分析。对Hub节点基因在20例OLP患者和20例健康对照者外周血中的表达进行检测。结果:在OLP与健康对照组之间共有212个差异表达基因。在构建的蛋白质网络中,所识别的Hub节点为GNB2L1,其在OLP患者外周血的表达显著高于健康对照组(P<0.001)。富集分析结果显示,212差异表达基因富集的通路有29个。所富集的细胞生物学过程有:信号转导、免疫反应、细胞通讯,类固醇代谢、突触小泡转运等。结论:本研究所识别出来的29条通路及Hub节点GNB2L1可能在OLP发生发展中起到关键作用。
Objective:To excavate and identify the biological pathways and the key genes or molecular for oral lichen planus(OLP)by using the bioinformatics methods.Methods:Gene expression profile GSE38616 was obtained from GEO database.Differentially expressed genes were screened by T test and Benjamini-Hochberg method,and then protein interaction network was constructed.The Hub node was identified by using Cytoscape plugin.Finally,the differentially expressed genes were enriched for pathway analysis.In addition,the expression of the Hub gene in peripheral blood of 20 OLP patients and 20 healthy controls was detected.Results:A total of 212 differentially expressed genes were selected between health and illness gingival tissue(P<0.01).In the construction of protein network,a hub node(RACK1,receptor for activated C kinase 1)was dug out.RACK1 expression in peripheral blood of OLP patients was significantly higher than that of healthy controls(P<0.001).Enrichment analysis showed that 29 pathways were enriched.And the enrichment of cell biology process including signal transduction,immune response,cell communication,steroid metabolism and synaptic vesicle transport.Conclusion:By using the bioinformatics methods,29 pathways and one hub molecular RACK1 may have important roles in the development of OLP.
