大黄总蒽醌超声辅助提取工艺优化及抑菌活性研究 被引量:4

Optimization in Ultrasonic-Assisted Extraction Process of Total Anthraquinone from Radix et Rhizoma Rhei and Its Antibacterial Activity

摘要 以大黄总蒽醌含量为评价指标,通过单因素实验和正交实验对大黄总蒽醌的超声辅助提取工艺进行了优化,并对其抑菌活性进行了研究。结果表明,在液料比为40∶1(mL∶g)、乙醇体积分数为90%、超声时间为35min、超声温度为70℃的最优提取工艺条件下,所得大黄总蒽醌含量达19.005mg·g^(-1);抑菌实验表明,大黄总蒽醌的抑菌活性随其浓度的增加逐渐增强,其对金黄色葡萄球菌和大肠杆菌均具有明显的抑制作用。超声辅助法提取大黄总蒽醌具有节省时间、方便快捷等优点。 Using the content of total anthraquinone from Radix et Rhizoma Rhei as an evaluation index,we optimized the ultrasonic- assisted process of total anthraquinone from Radix et Rhizoma Rhei by single-factor experiments and orthogonal experiments,and studied its antibacterial activity.The results show that the content of total anthraquinone from Radix et Rhizoma Rhei is up to 19.005 mg·g^-1 under the optimum extraction conditions as follows:the liquid- solid ratio of 40∶1(mL∶g),the ethanol volume fraction of 90%,the ultrasonic time of 35 min,and the ultrasonic temperature of 70 ℃.Moreover,the results of antibacterial experiments show that the antibacterial activity of total anthraquinone from Radix et Rhizoma Rhei increases with the increase of content.The total anthraquinone from Radix et Rhizoma Rhei has strong antibacterial activity against Staphylococcus aureus and Escherichia coli .The ultrasonic-assisted extraction process of total anthraquinone from Radix et Rhizoma Rhei has some advantages such as time saving and convenience.
