高糖环境下血管紧张素Ⅱ对肾小球系膜细胞生长影响的研究 被引量:1

摘要 目的探讨高糖培养条件下血管紧张素Ⅱ对肾小球系膜细胞(HMC)生长的影响。方法培养HMC,分为对照组、高糖模型组、高糖模型+血管紧张素Ⅱ组。高糖模型组在DMEM培养液中加入葡萄糖,终浓度为30 mmol/L;高糖模型+血管紧张素Ⅱ组在高糖模型中加入血管紧张素至浓度为10^(-6) mol/mL。MTT检测细胞增殖;流式检测细胞凋亡;Western Blot和荧光定量PCR检测核转录因子KappaB(NF-κB)、信号传导及转录激活因子3(STAT3)的表达。结果 NF-κB、STAT3的表达量,高糖模型+血管紧张素Ⅱ组最高,高糖模型组高于对照组;高糖环境下,10^(-6) mol/mL浓度的血管紧张素Ⅱ抑制了HMC增殖;细胞凋亡率,高糖模型组最高,对照组次之,高糖模型+血管紧张素Ⅱ组最低。结论高糖环境下血管紧张素Ⅱ会促使STAT3和NF-κB上调表达,激活JAK/STAT信号通路和炎症反应,但在一定浓度时会抑制HMC的增殖。
