小麦RPD3/HDA1基因家族的全基因组鉴定及其对热胁迫的响应 被引量:3

摘要 RPD3/HDA1是组蛋白去乙酰化酶(histone deacetylase,HDACs)家族中最大的亚族,已知的HDAC家族基因在拟南芥、水稻等植物的生长发育和非生物胁迫方面都发挥着重要作用,但小麦中对HDACs的研究却较少。为探究小麦中 RPD3/HDA1家族基因对热胁迫的响应,本试验利用生物信息学方法,从小麦全基因组中鉴定出36个小麦 RPD3/HDA1基因,并对其基因结构、进化关系、顺式作用元件和热胁迫下的表达模式进行分析。结果表明:除3A和4A外,其余小麦染色体均含有该家族基因,其基因结构中外显子和内含子的数目差异明显。该小麦家族基因在不同组织和不同逆境胁迫中存在差异表达, TaHDAC-Ⅱ-3a、 TaHDAC-Ⅱ-3b和 TaHDAC-Ⅱ-3d在叶片中的表达量明显高于其他组织, TaHDAC-Ⅰ-2d、 TaHDAC-Ⅰ-2a和 TaHDAC-Ⅰ-2b在热胁迫后表达量有明显上调。进一步研究发现,不同基因在同一生育时期的热胁迫下响应不同,同一基因在不同生育时期对热胁迫的响应也不同,并且相对于35℃而言,多数基因对42℃胁迫响应更显著。这些结果对寻找小麦 RPD3/HDA1家族候选耐热基因具有重要的参考价值。 RPD3/HDA1 is the largest subfamily of the histone deacetylase(HDACs)family.The known HDAC family genes play an important role in the growth and abiotic stress of Arabidopsis and rice.However,there are few studies on HDACs in wheat.To explore the response of theRPD3/HDA1 family of genes to heat stress in wheat,we identified 36 wheatRPD3/HDA1 genes from the whole genome of wheat using bioinformatics methods.The gene structure,evolutionary relationship,cis-acting elements and expression patterns under heat stress were analyzed.The results showed that except for 3A and 4A,the other wheat chromosomes contained the family gene,and the number of exons and introns in the gene structure was significantly different.The wheat family genes were deferentially expressed in different tissues and different stresses.The expression levels ofTaHDAC-Ⅱ-3a,TaHDAC-Ⅱ-3b andTaHDAC-Ⅱ-3d in leaves were significantly higher than those in other tissues,TaHDAC-Ⅰ-2d,TaHDAC-Ⅰ-2a andTaHDAC-Ⅰ-2b were significantly up-regulated after heat stress.Further studies had found that different genes responded differently under heat stress at the same growth period,and the same gene responded differently to heat stress at different growth stages.And most genes responded more significantly to stress at 42℃relative to 35℃.These results are important reference value for finding heat-resistant genes candidate of wheatRPD3/HDA1 family.
