益气活血方对冠心病大鼠药物作用靶点及治疗机制分析

摘要 目的:分析益气活血方对冠心病大鼠药物作用靶点及治疗机制。方法:选取从本院实验动物中心饲养的SPF健康雄性Wistar大鼠60只,将其随机分成三组,即对照组、假手术组与手术组,后两组大鼠均腹腔注射升压药物(左旋硝基精氨酸),制备冠心病大鼠模型。以不同浓度(低、中、高)的益气活血方以及盐酸地尔硫卓喂食大鼠,观察两组大鼠血浆中儿茶酚胺类物质去甲状肾上腺素、肾上腺素以及环磷酸腺苷的含量变化。结果:称重后发现对照组大鼠体重明显高于建立模型后大鼠,差异有统计学意义(P<0.05),而假手术组大鼠的体重与对照组相比差异无统计学意义(P>0.05);模型组大鼠的心脏体重比明显高于对照组、假手术组与中药高浓度组,且最大,与这三组的心脏体重比相比,差异有有统计学意义(P<0.05),与西药组、中药中浓度、低浓度组大鼠心脏体重比相比,差异无统计学意义(P>0.05),中药中浓度、低浓度组大鼠心脏体重比高于中药高浓度组大鼠,但与其心脏体重比相比,差异无统计学意义(P>0.05);模型组大鼠NE、E、cAMP水平相比于对照组均有所增加,差异有统计学意义(P<0.05),中药高浓度组大鼠NE、E、cAMP水平相比于模型组有所降低,差异有统计学意义(P<0.05),西药组大鼠NE、E、cAMP水平与模型组相比,差异无统计学意义(P>0.05),中药低浓度组大鼠NE、E、cAMP水平与中浓度组相比,差异无统计学意义(P>0.05),中药高浓度组与假手术组大鼠NE、E、cAMP水平与对照组相比,差异无统计学意义(P>0.05)。结论:益气活血方可调节大鼠血浆中儿茶酚胺类物质去甲状肾上腺素、肾上腺素以及环磷酸腺苷水平,增强心功能。 Objective: To analyze the therapeutic targets and therapeutic mechanism of Yiqi Huoxue Recipe on rats with coronary heart disease. Methods: Sixty healthy SPF male Wistar rats were selected from our hospital and randomly divided into three groups. The controlled group,the sham operation group,and the operation group were injected intraperitoneally with booster drugs(L Nitroarginine) was prepared in a rat model of coronary heart disease. The rats were fed with different concentrations(low,medium,and high) of Yiqi Huoxue Recipe and diltiazem hydrochloride. The contents of catecholamines,norepinephrine,epinephrine,and cyclic adenosine monophosphate in the plasma of rats were observed. Results: After weighing,it was found that the body weight(P<0.05). There was no significant difference in the body weight between the sham group and the controlled group(P<0.05). Heart and body weight ratio of the model group rats was significantly higher than the controlled group,the sham group and the high concentration group of traditional Chinese medicine,and also the largest,and compared with the three groups of heart and body weight ratio,there was statistically significant difference(P<0.05). There was no statistically significant difference(P>0.05) between heart weight ratios of rats in the western medicine group,Chinese herbal medicine medium concentration,and the low concentration groups. There was no significant difference in the concentration of rats in the concentration group compared with the heart weight ratio(P>0.05). The levels of NE,E,and cAMP in the model group were increased compared to the controlled group(P<0.05),NE,E,and cAMP levels in the high-concentration Chinese medicine group were lower than those in the model group(P<0.05). NE,E,and cAMP levels in the western medicine group were similar to those in the model group. There was no significant difference in the levels of NE,E and cAMP between the low-dose group and the middle-dose group(P>0.05). The NE,E,and cAMP levels in the high-dose and sham-operated groups of the rats were comparable to the controlled group. The ratio was not statistically significant(P>0.05). Conclusion: Yiqi Huoxue Recipe can regulate the level of catecholamines,epinephrine,epinephrine,and cyclic adenosine monophosphate in rat plasma and enhance cardiac function.
