蒙药材小黄紫堇的生药学鉴别和质量标准研究

Pharmacognosy identification and quality standard of monglia medicine Corydalis raddeana Regel

目的通过高效液相色谱法测定小黄紫堇中原阿片碱的含量,初步确立内蒙地区蒙药材小黄紫堇的质量标准。方法通过植物基源鉴别,性状鉴别及显微鉴别,对内蒙地区小黄紫堇进行生药鉴别研究,建立了内蒙地区小黄紫堇的薄层鉴别法和高效液相色谱法,确定了其药材中原阿片碱的含量。结果小黄紫堇药材的薄层色谱中显现明显的原阿片碱斑点,表明含此生物碱,高效液相法中对照品原阿片碱在81.6500~7.8979μg/mL范围内成良好的线性关系,准确度和精密度均良好,平均加样回收率为100.4%,RSD%=2.2%,七批次小黄紫堇中原阿片的含量为1.729~2.041mg/g。结论本研究所用方法操作简便、结果准确、重复性好,可作为内蒙地区小黄紫堇的生药鉴别和质量控制方法,为蒙药材小黄紫堇进一步药理研究提供了科学依据。

Objective Determination of the content of Protopine in Corydalis raddeana Regel by HPLC and the quality standard of Corydalis raddeana Regel of Mongolian medicinal herbs in Inner Mongolia region was preliminarily established. Methods Pharmacognosy identification of Corydalis raddeana Regel in Inner Mongolia were carried out by identification of plant basic source, identification of characters and microscopic identification. Then, the TLC and HPLC methods of Corydalis raddeana Regel in Inner Mongolia were established to determine the content of protopine. Results The obviously yellow spots of Corydalis raddeana Regel appeared in TLC chromatography indicated that medicinal plant contain protopine. A good linear relationship in the range of 81.6500~7.8979 μg/mL of the standard protopine was established. Average recovery of the spiking samples was 100.4%, RSD%=2.2%, the content of protopine in 7 batches of Corydalis raddeana Regel was 1.729~2.041 mg/g. Conclusion The methods in our study were simple, accurate and reproducible, and can be used as a method for identification and quality control of Corydalis raddeana Regel in Inner Mongolia, which provides scientific basis for further pharmacological study of Corydalis raddeana Regel.
