理气通便法治疗大鼠便秘合并痤疮的实验研究

Therapy of regulating qi and relaxing the bowels for treating constipation complicated with acne in rats:an experimental study

目的通过观察便秘合并痤疮大鼠模型血清白细胞介素(interleukin,IL)及肿瘤坏死因子α(tumor necrosis factor-α,TNF-α)的变化,分析理气通便法对免疫平衡相关因子的影响,探讨理气通便法治疗便秘合并痤疮的可能作用分子机制。方法采用99%油酸涂抹大鼠耳廓内部及75%次碳酸铋灌胃复制便秘合并痤疮模型大鼠,通过观察大鼠耳廓组织病理切片及粪便数量、质量及形态判定造模成功后,采用理气通便的中药治疗,并与西药莫沙必利片对照,酶联免疫法(ELISA)检测大鼠血清IL-1α、IL-2、IL-6、IL-10及TNF-α表达水平。结果与正常组比较,模型组大鼠血清IL-1α、IL-2、IL-6、TNF-α表达显著升高(P<0.05),IL-10表达显著下调(P<0.05);与模型组比较,各治疗组大鼠血清IL-1α、IL-2、IL-6、TNF-α表达显著下调(P<0.05),IL-10表达显著升高(P<0.05)。结论理气通便法治疗便秘合并痤疮可能与调控血液中炎症细胞因子水平,改善免疫紊乱有关。

Objective By observing the changes of serum interleukin (IL) and tumor necrosis factor-α(TNF-α) in the constipation complicated with acne rat model, to analyze the effects of regulating qi and relaxing the bowels on the immune balance related factors, and to explore the possible molecular mechanism of the treatment of constipation complicated with acne. Methods 99% oleic acid was applied to the rat's auricle and 75% bismuth subcarbonate by gavage to replicate the constipation complicated with acne model rats. The pathological sections of the auricle of the auricle and the quantity, quality and shape of the stool were observed to determine the establishment of model. The traditional Chinese medicine of regulating qi and relaxing the bowels were used to treat the rat model and compared with the western medicine Mosapride tablets. The levels of IL-1α, IL-2, IL-6, IL-10 and TNF-α in serum of rats were detected by ELISA. Results Compared with normal group, the expression of serum IL-1α, IL-2, IL-6, TNF-α in the model group increased significantly ( P <0.05) and the expression of IL-10 was significantly decreased ( P <0.05 ). Compared with model group, the expression of IL-1α, IL-6, IL-6 and TNF-α was significantly decreased ( P <0.05) in the treatment group, and the expression of IL-10 increased significantly ( P <0.05 ). Conclusion The treatment of constipation complicated with acne by regulating qi and relaxing the bowels may be related to regulating the levels of inflammatory factors and improving immune disorders.
