Optimization of Ethanol Extraction Process of Shanhuang Qutong Spray by Orthogonal Test

目的优选山黄祛痛喷雾剂醇提工艺。方法采用L9(34)正交试验法考察乙醇浓度、乙醇用量、回流时间、回流次数4个因素在不同水平对秋水仙碱、槲皮素含量和出膏率的影响。结果影响因素按程度大小依次为回流次数、乙醇浓度、回流时间和乙醇用量;山黄祛痛喷雾剂最佳醇提工艺为丽江山慈菇和凤仙透骨草采用8倍量70%乙醇,回流提取2次,每次1. 5 h。结论该醇提工艺稳定可行、质量可控,可用于山黄祛痛喷雾剂的制备。

Objective To optimize the ethanol extraction process of Shanhuang Qutong Spray.Methods L9(3^4)orthogonal test was used to investigate the effects of different levels of ethanol concentration,ethanol consumption,reflux time and reflux times on the content and the extraction rate of colchicine and quercetin.Results The influencing factors were reflux times,ethanol concentration,reflux time and ethanol consumption in order of the influence level.The optimum ethanol extraction process of Shanhuang Qutong Spray was as follows:Iphigenia indica and Impatiens balsamina were extracted with 8 times of 70%ethanol,reflux extraction for 2 times,and 1.5 h each time.Conclusion The ethanol extraction process is stable and feasible,the quality is controllable.It can be used for the preparation of Shanhuang Qutong Spray.
