摘要
目的探究不同pH溶剂对5-羟甲基糠醛(5-HMF)溶出效率的影响,优化生脉散的提取工艺。方法用高效液相色谱法(HPLC)测定5-HMF的含量为指标,采用单因素考察法确定最佳提取溶剂;采用正交试验法优化生脉散最佳煎煮工艺。结果以5%醋酸为溶剂时5-HMF的溶出率比水更好;生脉散最佳煎煮工艺为加10倍量5%醋酸,浸泡60min,煎煮1.5h,煎煮两次。结论酸性溶剂有利于生脉散有效成分5-HMF的形成。
Objective To investigating the effect of pH value on dissolution efficiency of 5-hydroxymethy-2-furaldehyde(5-HMF) and to optimize the extraction process of Shengmaisan. Methods The content of 5-HMF was determined by high performance liquid chromatography(HPLC).The optimum extraction solvent was determined by single factor method and the best decocted process of Shengmaisan was optimized by orthogonal test.Results When 5% acetic acid was used as solvent,the dissolution rate of 5-HMF was better than that of water.The optimum decoction technology of Shengmaisan was to add 10 times of 5% acetic acid,soaked for 60 minutes,and cooked 2 times,1.5 h each time.Conclusion The acidic solvent is beneficial to the formation of 5-HMF,an active ingredient of Shengmaisan.
