8个马大杂种相思无性系组培快繁试验初报 被引量:4

摘要 通过对马大杂种相思8个无性系号(10#、73#、LK1、LK2、LK3、SK1、SK2、SK3)的组培快繁进行试验,从影响外植体诱导、增殖、瓶内生根、驯化与移栽等因素进行研究。结果表明:马大杂种相思理想的外植体材料是当年生枝条第3~5个腋芽茎段;外植体两种消毒方法(70%酒精+0.1%升汞)和(2%次氯酸钠+0.1%升汞)对消毒效果没多大区别;两种消毒剂同时使用时务必掌握好消毒时间,0.1%升汞6min是最合适的,诱导率达58.2%;增殖效果与6 BA浓度有关,固定NAA为0.1mg·L-1,6BA最佳浓度以0.4mg/L和0.6mg/L为较适范围,同时与基本培养基有关,其中MS培养基对促进芽分化效果最显著,培养4个月时已进入旺盛增殖期,6个月的倍率为3.87;瓶内生根培养以V6:1/2MS+IBA1.2 mg/L+NAA0.2 mg/L和V13:1/2MS+NAA 1.2 mg/L+IBA0.2 mg/L的生根效果最好,生根率都达96%,生根数平均达3.6条以上;移栽基质选择以黄心土:沙为1:1效果最合适的;8个号移栽的平均成活率有一定的差异,超过80%是10#(87.3%)、73#(83.5%)、SK1(87.8%),其它号为70%左右;制定了马大相思从增殖倍率、生根率及瓶内生根生产能力的综合评价体系,初步认定SK1、10#、73#3个号为可形成瓶内生根生产能力并规模工厂化生产苗木的品号。 We test the histological propagation of 8 asexual numbers(10 #,73 #,LK1,LK2,LK3, SK1,SK2,SK3)of the Acacia Mongrel. The effects of induction,proliferation, rooting,domestication and transplanting were studied. The results showed that the ideal exophytic material of Acacia was the 3rd to 5th axillary shoots of the same year. The two disinfection methods for implants(70 % alcohol + 0.1 % Mercury)and(2 % sodium hypochlorite + 0.1 % Mercury)have little difference in the disinfection effect,and the success rate is more than 30 %. When two disinfectants are used at the same time,must master the disinfection time. 0.1 % litres of mercury for 6 minutes is the most appropriate,with an induction rate of 58.2 %;The proliferation effect is related to the 6 BA concentration. The fixed NAA is 0.1 mg·L^-1. The best concentration of 6 BA is 0.4mg/L和0.6mg/L,and it is related to the basic medium. The MS medium has the most significant effect on promoting bud differentiation. At the time of cultivation for 4 months, it has entered a period of vigorous proliferation,and the doubling rate for 6 months is 3.87;The rooting effect of V6:1 / 2MS + IBA1 .2 mg/L + NAA 0.2 mg/L and V13:1 / 2MS + NAA 1.2 mg/L + IBA 0.2 mg/L was best in bottle rooting culture. The rooting rate reached 96 %, and the average number of roots reached 3.6 or more;Transplantation matrix selection with LOESS soil: sand is 1:1 effect is appropriate;The average survival rate of transplanting is 8 with certain differences, with more than 80 % being 10 #(87.3 %),73 #(83.5 %),SK1(87.8 %),and other numbers being about 70 %;A comprehensive evaluation system of the multiplication rate,rooting rate,and rooting capacity of Acacia matha was established,and SK1,10 #,73 # 3 numbers were initially identified as the numbers that could form the rooting capacity in the bottle and scale the factory production of seedlings.
