p75NTR对小鼠坐骨神经挤压伤后神经功能的影响 被引量:2

THE EFFECT OF p75^(NTR) ON THE NERVE FUNCTION OF MICE AFTER THE SCIATIC NERVE CRUSH INJURY

摘要 目的探讨p75^(NTR)对小鼠坐骨神经挤压伤后神经功能的影响。方法将20只野生型C57BL/6雄性小鼠分为模型组和假手术组,每组各10只;选取10只p75^(NTR)基因敲除雄性小鼠为p75^(NTR-/-)组。p75^(NTR-/-)组和模型组小鼠钳夹左侧坐骨神经,制作神经挤压伤模型;假手术组小鼠仅分离、暴露左侧坐骨神经。观察术前和术后不同时间点3组小鼠SFI、趾展试验及斜板试验等行为学变化。结果成模第7、14、21、28、35天,p75^(NTR-/-)组SFI均低于模型组(P<0.05);p75^(NTR-/-)组趾展试验初始反应时间及完全反应时间较模型组明显延长(P<0.01);在成模第7、14、21、28、35天,p75^(NTR-/-)组斜板试验角度均低于模型组(P<0.05)。结论 p75^(NTR)对小鼠坐骨神经损伤后神经功能恢复发挥重要作用。 Objective To investigate the effect of p75NTR on the nerve function of mice by observing the behavioral function changes in p75 NTR gene knockout mice and wild-type C57BL/6 mice after sciatic nerve crush injury. Methods 20 wild-type C57BL/6 male mice were randomly divided into model group and sham group with 10 mice in each group. 10 p75^NTR-/- male mice with a C57BL/6 background were selected as p75NTR/group. The mice of the model group and p75^NTR-/- group were made as left sciatic nerve crush injury models. In the sham group, only left sciatic nerve exploration was carried out without nerve being crushed. Some behavioral methods such as SFI, toe spreading test and inclined board test were measured respectively during pre-operation and at different time points during post-operation. Results The SFI of the model group was higher than that of p75^NTR-/- group on day 7,day 14, day 21, day 28 and day 35 after injury ( P <0.05). The initial reaction time and complete reaction time of p75^NTR-/- group were significantly longer than that of model group ( P <0.01). The angle of p75^NTR-/- group was much lower than that of the model group on day 7,day 14, day 21, day 28, and day 35 after injury ( P <0.05). Conclusion p75NTR played a significant role during the recovery of sciatic nerve crush injury.
