摘要
为探究川续断水煎剂对小鼠血清中免疫球蛋白(lmmunoglobulim,lg)和细胞因子的影响,评估其对动物机体免疫功能的调节作用。将40只昆明系健康小鼠,雌雄各半,随机分为5组,每组8只,分别按体质量灌胃10 g/kg·bw、20 g/kg·bw、30 g/kg·bw、40 g/kg·bw终浓度为2 g/mL的川续断水煎剂和生理盐水,连续给药7 d。于末次给药后第7天无菌操作摘除眼球取血分离血清,测定小鼠血清中免疫球蛋白和细胞因子水平。结果显示,与对照组相比,灌服不同剂量的川续断水煎剂均能显著提高小鼠血清中IgA和IgM水平(P<0.05),不同程度地提高IgG的水平;显著升高小鼠血清中IL-2、INF-γ和TNF-α水平(P<0.05),显著降低IL-4和IL-10水平(P<0.05)。表明川续断水煎剂可升高小鼠血清中免疫球蛋白水平,同时能增强血清中Th1类细胞因子水平,降低Th2类细胞因子的水平,提示川续断可增强动物机体的非特异性免疫功能。
This study was conducted to investigate the effect of decoction of Radix Dipsaci on serum immunoglobulin and cytokines in mice.Forty Kunming mice with half males and half females were randomly divided into 5 groups with 8 mice in each group.All the experimental mice were intragastrically administrated decoction of Radix Dipsaci(10,20,30 and 40 g/kg·body weight,respectively)with the final concentration of 2 g/mL and physiological saline for 7 d,respectively.Blood samples were sterilely collected from eyeball seven days after the last administration.And the immunoglobulin and cytokines levels in serum were detected.The results showed that the levels of IgA and IgM in serum were significantly increased after oral administration of decoction of Radix Dipsaci compared with control group(P<0.05),and tended to enhance serum IgG level.Oral administration of decoction of Radix Dipsaci also significantly enhanced serum IL-2,INF-γand TNF-αlevels(P<0.05)and also significantly reduced serum IL-4 and IL-10 levels(P<0.05).In conclusion,the decoction of Radix Dipsaci can increase immunoglobulin levels in serum of mice,and enhance the level of Th1 type cytokines and reduce the level of Th2 type cytokines.Our findings suggest that Radix Dipsaci strengthens the nonspecific immune function of animals.
