何首乌饮对离体大鼠睾丸间质细胞p53、Bcl2和BAX mRNA表达的影响 被引量:5

摘要 目的:探讨何首乌饮对离体大鼠睾丸间质细胞p53、 Bcl2、BAX mRNA表达的影响。方法:用自由基氧化损伤方法建立睾丸间质细胞衰老模型,通过β-半乳糖苷酶染色观察何首乌饮对离体大鼠睾丸间质细胞衰老情况的影响;应用实时荧光定量PCR检测p53、Bcl2和BAX mRNA在间质细胞中表达水平。结果:间质细胞模型组β-半乳糖甘酶含量明显高于正常组,何首乌饮能明显降低间质细胞衰老标志物β-半乳糖甘酶的含量。模型组间质细胞p53、BAX的mRNA水平明显高于正常组,Bcl2的mRNA表达水平低于正常组;何首乌饮干预组的p53、BAX的mRNA表达低于模型组,Bcl2的mRNA表达水平高于模型组。结论:何首乌饮可以上调间质细胞Bcl2mRNA表达水平,下调p53和BAX的mRNA表达。 Objective : To investigate the effect of Heshouwuyin on the mRNA expression of p53, Bcl2 and BAX in testicular Leydig cells of rat in vitro. Methods : Free-radical oxidative damage method was used to establish the cell aging model. The influence of Heshouwuyin in Leydig cells of isolated rat was observed by β-galactosidase staining.Real-time PCR was used to detect the mRNA expression of p53, Bcl2 and BAX. Results: The content of β-galactosidase in the Leydig cells of the model aging group was significantly increased compared with the normal control group.Heshouwuyin could significantly reduce the content of β-galactosidase regarded as aging relevant marker in Leydig cells. Compared with the normal control group, the p53 and BAX mRNA expression levels of the aging group were significantly increased, and the Bcl2 mRNA expression level was significantly decreased. Compared with the aging group, the p53 and BAX mRNA expression levels of the Heshouwu decoction group were significantly lower, and the Bcl2 mRNA expression level was significantly increased. Conclusion : Heshouwu decoction can up-regulate the expression of Bcl2 mRNA in Leydig cells and inhibit the mRNA expression of p53 and BAX. Heshouwu decoction can down-regulate the apoptosis of rat Leydig cells and delay the aging of cells.
